miR-21与miR-375在维吾尔族、哈萨克族及汉族食管鳞癌患者组织和血浆中的表达差异

【摘要】:目的:探讨微小RNA-21、微小RNA-375在维吾尔族、哈萨克族及汉族食管鳞状细胞癌患者组织及血浆中的表达情况及发病风险。方法:选取180例食管鳞癌患者,并经术后病理确诊,术前采集其血液标本,术后分别采集食管癌及癌旁组织,实验方法采用实时荧光定量PCR法检测hsa-miR-21-5p、hsa-miR-375在组织及血液标本中的表达情况;应用配对样本的卡方检验分析癌和癌旁组织中miR-21和miR-375的表达差异;应用单因素方差分析血浆中miR-21和miR-375在不同民族间的表达水平。结果:miR-21在食管组织中表达水平较癌旁组织高(P0.05),食管鳞癌组织中miR-375表达水平较癌旁组织低(P0.05),但3个民族间无明显差异;哈萨克族和维吾尔族食管鳞癌患者血浆中miR-21表达水平较汉族高(P0.05);miR-21高表达及miR-375的低表达增加了食管癌的发病风险,可能是影响食管癌的危险因素。结论:miR-21、miR-375参与食管鳞状细胞癌的发生、发展,miR-21、miR-375或可成为食管癌早期诊断的潜在肿瘤标志物。
[Abstract]:Objective: to investigate the expression and risk of microRNA-21 and RNA-375 in tissues and plasma of Uygur, Kazak and Han patients with esophageal squamous cell carcinoma.Methods: 180 cases of esophageal squamous cell carcinoma were selected and confirmed by pathology. Blood samples were collected before operation, and esophageal carcinoma and adjacent tissues were collected respectively after operation.Methods the expression of hsa-miR-21-5phsa-miR-375 in tissues and blood samples was detected by real-time fluorescence quantitative PCR, and the difference between miR-21 and miR-375 expression in cancer and adjacent tissues was analyzed by chi-square test.Univariate ANOVA was used to analyze the expression of miR-21 and miR-375 in different ethnic groups.Results the expression of miR-375 in esophageal squamous cell carcinoma was lower than that in paracancerous tissue (P 0.05), but there was no significant difference among the three ethnic groups.The higher expression of miR-21 and the lower expression of miR-375 in the plasma of Kazak and Uygur patients with esophageal squamous cell carcinoma increased the risk of esophageal cancer, which may be the risk factors of esophageal cancer.Conclusion miR-21 miR-375 may be involved in the development of esophageal squamous cell carcinoma and may be a potential tumor marker for early diagnosis of esophageal carcinoma.
