DIAPH3蛋白对三阴性乳腺癌细胞增殖和迁移侵袭能力的影响
[Abstract]:Objective: to investigate the effect of DIAPH3 protein on the migration and invasion of triple-negative breast cancer cells. Methods: to investigate the expression of DIAPH3 protein in MCF-10A,SUM159,MCF-7 and MDA-MB-231 cells by Q PCR and western blot. Then the overexpression vector DIAPH3, was constructed to analyze the effect of overexpression of DIAPH3 protein on the migration and invasion of triple-negative breast cancer cells by Transwell. Finally, the molecular mechanism of DIAPH3 protein affecting the migration and invasion of triple-negative breast cancer cells was discussed. Results the relative expression levels in MCF-7 and MDA-MB-231 were 1.00 卤0.000,0.47 卤0.068,0.64 卤0.039 and 0.17 卤0.008, respectively. Compared with normal breast epithelial cell MCF-10A, the relative expression of DIAPH3 protein in SUM159,MCF-7 and MDA-MB-231 of breast cancer cells was significantly lower than that of normal breast epithelial cells (P0.05). The relative expression of DIAPH3 protein in SUM159 and MDA-MB-231 was significantly lower than that in MCF-7 (P0.05). The number of migration of MDA-MB-231 cells in group,NC group and DIAPH3 group was 305.5 卤6.36329 卤16.97 and 221 卤24.04Cell group,NC group, respectively, and the number of invasion of MDA-MB-231 cells in DIAPH3 group was 96 卤8.25103 卤18.59 and 24.5 卤5.68, respectively, compared with Cell group,NC group. Overexpression of DIAPH3 could significantly inhibit the migration and invasion of MDA-MB-231 cells, and further studies showed that overexpression of DIAPH3 protein could inhibit the expression of Rho Agna MMP-2 and MMP-9 proteins. Conclusion the expression of DIAPH-3 in breast cancer cells was significantly lower than that in triple-negative breast cancer cells, and the overexpression of DIAPH-3 could inhibit the expression of MMP-2 and MMP-9 proteins in TNBC cells, thus inhibiting the migration and invasion of TNBC cells.
