胃腺癌血清非炎症性标志物的筛

研究背景胃腺癌是胃癌的一种,源于胃腺体细胞恶化,占全部胃癌发病率的95%。早期诊断可明显改善癌症治疗效果,但现实却是由于缺乏早期诊断手段,往往就诊时多数患者已经处于局部晚期或发生了远处转移从而延误了病情,错失最佳的治疗时机。血清肿瘤标志物以其简便、快捷、无痛苦等优点,成为胃癌患者的理想检查指标,目前临床常见的胃癌辅助诊断中有癌胚抗原(CEA)、糖类抗原125(CA125)及糖类抗原199(CA19-9)。尽管多种肿瘤标志物联合检测能够在一定程度上提高早期胃癌诊断的敏感度,可用于预测患者的病情进展和预后,但检测重复性不佳。组合应用表面增强激光解析电离飞行时间质谱(SELDI-TOF-MS)、高效液相色谱(HPLC)及基质辅助激光解析附电离飞行时间质谱(MALDI-TOF-MS)技术,课题组前期曾成功在乳头状甲状腺癌及乳腺癌中寻找特异性标志物,对相关肿瘤早期诊断及预后检测有参考意义,但前期实验存在以下问题:a)血清特异性标志物未与临床在使用的肿瘤标志物做对比;b)特异性标志物未排除炎症干扰;c)血清分析测试前各种影响因素存在偏差,且相互不独立,影响检验效能。尽管近年来分子生物标志物的发现急...
