ECRG4表达与胃癌临床病理学关系及相关机制的研究

目的:近年来恶性肿瘤的发生率和死亡率呈上升趋势,严重危害着人类健康。胃癌是我国、日本、韩国等东亚国家高发恶性肿瘤之一,胃癌的发生发展是个人、环境及其他多种因素、多阶段共同作用的结果,包括遗传学,表遗传学及环境因素等。表遗传学事件成为近些年研究的热点,通过影响基因转录活性抑制基因表达,却不涉及DNA序列的改变。表遗传学修饰包括DNA甲基化,组蛋白修饰,染色体重塑,RNA干扰等,其中DNA甲基化和组蛋白修饰两种表遗传学现象研究最为广泛。肿瘤抑制基因失活包括基因缺失,基因突变和DNA异常甲基化三种机制。研究显示胃癌发生、发展和形成过程中肿瘤抑制基因发生甲基化频繁出现。食管癌相关基因4(Esophageal Cancer Relaetd Gene4,ECRG4),是98年中国医学科学院Bi等利用m RNA差异显示技术从食管癌组织中分离得到的。该基因位于人染色体2q14.1-14.3,有4个外显子,全长12,500bp,其开放阅读框架为444bp,编码148个氨基酸多肽链,蛋白产物分子量为17k Da,在该基因的核心启动子区、第一外显子和第一内含子5′端的大约1kb的范围内分布有多个Cp G岛,...
