miR-501-3p在肾癌中的抑癌作用及其机制研究

背景:肾癌即肾细胞癌(renal cell carcinoma),是泌尿系统常见的恶性肿瘤之一,其起源于肾实质泌尿小管上皮系统,占肾脏恶性肿瘤的80%~90%,发病率约占全身肿瘤的2.1%。近年来由于吸烟、饮食变化等原因,肾癌发病率有逐渐上升的趋势。肾癌易进展,预后差,且其分子机制尚不明确。阐明肾癌发生发展的生物学机制,对于肾癌早期诊断及治疗具有重要的临床意义。microRNAs(miRNAs)是一类由内源性基因编码的非编码RNA,通过靶向基因mRNA3’端非编码区调控基因表达。研究表明miRNAs如miR-501-3p参与肿瘤的发生发展,过表达miR-501-3p可抑制前列腺癌细胞增殖和集落形成,并诱导细胞周期G1期阻滞。miR-501-3p在肺癌、大肠癌、胰腺导管腺癌等肿瘤中也有促癌或抑癌作用。但miR-501-3p在肾癌中的表达模式及其生物学功能仍缺乏系统性研究,其对肾癌细胞增殖、迁移的影响及其分子机制尚不明确。目的:通过生物信息学网站分析miR-501-3p在肾癌中的表达模式并分析其表达下调的表观遗传学机制。采用一系列功能实验探究miR-501-3p在肾癌中的生物学功能。利用生...
