“百会”透“曲鬓”对急性期脑出血大鼠脑组织中Survivin表达影响的实验研究

【摘要】：目的:采用"百会"透"曲鬓"穴的针刺方法,观察在针刺治疗过程中Survivin在脑出血后大鼠脑组织中的动态变化,观察Survivin与脑出血后细胞凋亡之间的关系,从而探讨其对急性期脑出血大鼠脑组织中Survivin表达的影响,为临床上治疗急性期脑出血提供有利的依据。方法:75只雄性Wistar大鼠,随机分为空白组、模型组和针刺组。其中空白组15只大鼠,模型组和针刺组各30只,每组又平均分为6h,12h,24h,3d,7d五个时间点。其中针刺组和模型组采用自体血注入法建立脑出血模型。待大鼠苏醒后观察并记录其神经功能缺损程度,挑选符合条件的模型。对针刺组进行针刺"百会"透"曲鬓"的治疗并对各组大鼠进行系统的实验观察:(1)观察各组大鼠神经功能情况;(2)采用免疫组化检测各组大鼠脑组织中Survivin的相关动态表达;(3)采用TUNNEL法检测各组大鼠脑出血后血肿周围细胞凋亡情况。结果:(1)神经功能评分结果:针刺组与模型组在12h、24h、3d的时间点比较有明显差异,可观察到针刺组神经功能缺损评分得到明显改善,有统计学意义(P0.01);(2)免疫组化结果显示:通过针刺组和模型组比较,针刺组Survivin在各个时间点均有动态表达,尤其是在12h、24h、3d表达明显增高(p0.01);(3)TUNEL法检测结果:针刺组与模型组比较,针刺组在各个时间点细胞凋亡数量均在减少,尤其是在12h、24h、3d时间点差异有统计学意义。(P0.01)。结论:(1)针刺"百会"透"区鬓"穴针刺法对脑出血后神经功能缺损有明显改善,具有神经保护作用;(2)针刺"百会"透"曲鬓"通过提高Survivin的表达,对脑出血后血肿周围脑组织起到保护作用;(3)针刺"百会"透"曲鬓"能够提高TUNEL的阳性表达,抑制细胞凋亡。
[Abstract]:Objective: using the "method of acupuncture of Baihui Qubin acupoint, observe the dynamic changes in the acupuncture treatment process of Survivin in rat brain after cerebral hemorrhage, the relationship between Survivin and apoptosis after intracerebral hemorrhage, and to explore its effect on the expression of Survivin in brain tissue of rats with acute cerebral bleeding, for the clinical treatment of acute cerebral hemorrhage with a favorable basis. Methods: 75 male Wistar rats were randomly divided into the blank group, the model group and the acupuncture group. There were 15 rats in the blank group, 30 in the model group and in the acupuncture group. Each group was divided into five time points of 6h, 12h, 24h, 3D and 7d. In the acupuncture group and the model group, the model of cerebral hemorrhage was established by injection of autologous blood. After the rats were awakened, the degree of nerve function defect was observed and recorded, and the model was selected. The acupuncture group acupuncture "Baihui" treatment through "Qubin" and the rats of each group were observed: (1) experimental system to observe the neural function of rats; (2) the relevant dynamic expression by using immunohistochemical Survivin was detected in the brain tissue of rats; (3) detected by TUNNEL the rats after ICH apoptosis. Results: (1) the nerve function score results: acupuncture group and model group in 12h, 24h, 3D time points, there was significant difference between the observed score, acupuncture group of neurological deficit improved significantly, with statistical significance (P0.01); (2) immunohistochemistry results showed that: the acupuncture group and model group comparison of expression of Survivin in the acupuncture group at each time point were dynamic, especially the expression is significantly increased in 12h, 24h, 3D (P0.01); (3) TUNEL test results: acupuncture group compared with model group, the acupuncture group at each time point the number of cell apoptosis was decreased, especially in 12h, were statistically significant 24h, 3D time points difference. (P0.01). Conclusion: (1) acupuncture "Baihui" through "temple area" point acupuncture can obviously improve the neurological function after cerebral hemorrhage, has a neuroprotective effect; (2) acupuncture "Baihui Qubin through improving the expression of Survivin, to protect the brain tissue around the hematoma after intracerebral hemorrhage; (3) acupuncture" Baihui Qubin can increase the expression of TUNEL, inhibit apoptosis.
