参地补肾胶囊对肾间质纤维化大鼠Snail及E-cadherin蛋白表达的影响

【摘要】:目的:通过动物实验研究,观察参地补肾胶囊对单侧输尿管梗阻(unilateal ureteral obstruction,UUO)所致肾间质纤维化大鼠模型肾功及Snail、E-cadherin蛋白表达的影响,探讨参地补肾胶囊保护肾功能、抗肾间质纤维化的作用机理。方法:将70只雄性SD大鼠随机分为空白组、假手术组、模型组、尿毒清组、参地补肾组,每组14只。采用单侧输尿管结扎的方法建立肾间质纤维化模型,证实造模成功后开始灌胃,尿毒清组给予尿毒清颗粒混悬液灌胃,参地补肾组给予参地补肾胶囊混悬液灌胃,空白组、假手术组、模型组均用等量生理盐水灌胃,连续灌胃28天。观察治疗前后大鼠的一般状态,Scr、BUN的变化;将肾组织病理切片行HE染色,观察肾脏病理学改变;采用免疫组化法检测肾组织中Snail、E-cadherin表达的情况。结果:参地补肾组大鼠的一般状态明显好于模型组;参地补肾组大鼠肾脏病理改变明显轻于模型组;参地补肾组大鼠在第35天血清Scr、BUN均明显降低,与模型组比较有统计学意义(P0.05);参地补肾组大鼠在第35天肾组织中Snail表达明显降低,E-cadherin表达明显升高,与模型组比较有统计学意义(P0.05)。结论:参地补肾胶囊在一定程度上改善UUO大鼠的一般状态,降低大鼠Scr、BUN水平;参地补肾胶囊能减轻UUO大鼠肾脏病理学改变;参地补肾胶囊可能通过下调Snail的表达,上调E-cadherin的表达,在一定程度上延缓肾间质纤维化的进展。
[Abstract]:Objective: through animal experiments, observe ginseng Bushen Capsule on unilateral ureteral obstruction (unilateal ureteral obstruction, UUO) induced renal interstitial fibrosis in rat model of renal function and the expression of Snail and E-cadherin protein of ginseng, kidney capsule protect renal function and antitumor mechanism of renal interstitial fibrosis. Methods: 70 male SD rats were randomly divided into the blank group, the sham operation group, the model group, the uremia group and the Shen Di kidney tonifying group, with 14 rats in each group. Methods unilateral ureteral ligation of renal interstitial fibrosis model, confirmed after the success of modeling to gavage, Niaoduqing group with Niaoduqing granule suspension gavage, ginseng ginseng Bushen group given Bushen capsule suspension gavage, blank group, sham operation group, model group with normal saline intragastric gavage for 28 days. The changes of general condition, Scr and BUN in rats before and after treatment were observed. The pathological changes of renal tissue were observed by HE staining, and the expression of Snail and E-cadherin in renal tissue was detected by immunohistochemistry. Results: the general state of ginseng kidney group rats was significantly better than the model group; ginseng kidney group rat kidney pathological changes were obviously less than that of the model group; ginseng kidney group rats after thirty-fifth days of serum Scr, BUN were significantly decreased, had statistical significance compared with the model group (P0.05); ginseng kidney group the rats in the thirty-fifth days of renal tissue Snail expression was significantly reduced, E-cadherin expression was significantly increased and was statistically significant compared with the model group (P0.05). Conclusion: Ginseng Bushen capsule improving the general condition of UUO rats to a certain extent, reduce the level of Scr and BUN rats; ginseng Bushen capsule can alleviate the pathological changes of renal pathology in UUO rats; ginseng Bushen capsule could downregulate the expression of Snail, increase the expression of E-cadherin and delay the progression of renal interstitial fibrosis in to a certain extent.
