壮药大叶金花草质量评价研究

【摘要】:目的:对壮药大叶金花草进行真伪、优劣评价,初步建立大叶金花草质量评价方法。方法:通过文献考证明确其历史沿革;利用原植物鉴定、性状鉴定与理化鉴定的方法进行生药学鉴定研究;以显微鉴定的方法鉴别大叶金花草与小金花草;按《中国药典》2015年版的方法进行药材水分、灰分、浸出物等检查项目相关研究;采用HPLC一测多评法,同时测定其中4种成分的含量;对其进行高效液相指纹图谱研究。结果:(1)明确大叶金花草的品种来源、功能效用,为大叶金花草的临床应用提供依据。(2)原植物形态、药材性状特征与薄层色谱均具有鉴别意义。(3)大叶金花草孢子为一侧凹陷的卵球形,根茎具有1个周韧型管状中柱,中央有髓,叶柄具1个“U”形维管束,叶表皮细胞壁不增厚;小金花草孢子为球形或类三角形的四面体,根茎分体中柱周韧型,叶柄具2个维管束,叶表皮细胞外壁略增厚。(4)大叶金花草药材根及根茎水分含量在6.64%~10.35%,总灰分含量在8.50%~15.91%,酸不溶性灰分在4.98%~11.10%,浸出物含量在7.95%~21.40%之间;全草药材含水量为7.45%~12.24%,总灰分为5.04%~11.29%,酸不溶性灰分范围为1.39%~6.58%,浸出物含量为24.96%~38.05%;地上部分水分含量幅度为8.35%~15.62%,总灰分含量幅度为3.94%~5.00%,酸不溶性灰分幅度为0.17%~0.80%,浸出物含量幅度为26.61%~35.76%。(5)一测多评所得的含量计算结果与外标法所得结果无显著性差异,在不同的实验条件下,各成分之间的校正因子重现性好(RSD3%),因此一测多评法可以应用于大叶金花草的质量评价。(6)建立了大叶金花草指纹图谱共有模式,标定了14个共有峰,确定了其中4个主要色谱峰所对应的物质。结论:以上实验及研究对壮药大叶金花草药材质量进行了初步评价,为其临床应用的安全、有效和可控提供参考。本论文研究内容经科技查新证实,目前尚未见与上述研究相同的国内外文献报道。
[Abstract]:Objective: to evaluate the true and false quality of Flos aurantia and to establish a method of quality evaluation. Methods: the historical evolution was confirmed by textual research. The methods of original plant identification, character identification and physicochemical identification were used to identify the pharmacognosy. The method of microscopical identification was used to identify Amomum macrophylla from Flos aurantia. According to the method of 2015 edition of Chinese Pharmacopoeia, the related studies on water, ash and extract of medicinal materials were carried out. The content of four components was determined by HPLC method. The results showed that the origin and function of the species were determined, which provided the basis for clinical application. The characteristics of medicinal materials and TLC have the distinguishing significance. 3) the spores are ovoid in one side of the sag, the rhizome has a round and tough tubular column, and the center is myelinated. The petiole has a "U" vascular bundle, and the cell wall of the leaf epidermis is not thickened. The spores are globose or triangular tetrahedron, rhizomatous in the column round tough type, petiole with two vascular bundles. The content of water in the root and rhizome of Phyllostachys davidii was 6.64 and 10.35, and the total ash content was 8.500.91%. The acid insoluble ash content was 4.98% and 11.10%, and the extract content was between 7.95% and 21.40%. The water content of the whole herb is 7.450.12.24, the total ash is 5.0411.29 and the acid insoluble ash is 1.396.58%. The content of the extract was 24.96% and 38.05%; The water content of the aboveground part was 8.35 and 15.62, the total ash content was 3.94 and 5.00, the acid insoluble ash was 0.17% and 0.80%. There was no significant difference between the calculated results of multi-evaluation and that of external standard method, and under different experimental conditions, the content of extracts was 26.61% and 35.76% (5%), and there was no significant difference between the calculated results and the results obtained by external standard method. The reproducibility of the calibration factors among the components is good. Therefore, a method of multiple evaluation can be applied to the quality evaluation of Flower and Flower. 6) A common pattern of fingerprint of Flower and Flower is established. The 14 common peaks were calibrated, and the corresponding substances of four main chromatographic peaks were determined. Conclusion: the quality of medicinal materials of Chlorophyllum grandiflorum was preliminarily evaluated by the experiments and studies above, which is the safety of its clinical application. The research content of this paper has been confirmed by scientific and technological review, and there is no domestic and foreign literature report which is the same as the above research at present.
