砂地柏总黄酮抗炎镇痛及免疫调节作用实验研究

【摘要】:目的:研究维吾尔药砂地柏总黄酮(JSTF)的抗炎镇痛及免疫调节作用,为其深入开发提供基础数据。方法:(1)采用二甲苯致小鼠耳肿胀、小鼠毛细血管通透性、大鼠棉球肉芽肿以及蛋清、组胺和角叉菜胶致大鼠足趾肿胀实验,以阿司匹林为阳性对照,观察JSTF对炎症的抑制作用;(2)采用冰醋酸致小鼠扭体法、热板法和甲醛致小鼠疼痛实验,观察JSTF的镇痛作用;(3)分别采用碳粒廓清和血清溶血素测定实验观察JSTF对小鼠非特异性和特异性体液免疫功能的影响。结果:(1)低、中、高剂量组的JFST(125、250、500 mg/kg)能明显抑制二甲苯致小鼠耳廓肿胀的发生(P0.05),其抑制率分别为(16.22%,40.67%,51.78%)。低、中、高剂量组的JFST(125、250、500 mg/kg)也能明显抑制小鼠毛细血管通透性的发生(P0.05),其抑制率分别为(11.63%,32.56%,53.49%)。在角叉菜胶引起大鼠足趾肿胀实验中,除了第一个时间段内JSTF低剂量组和模型组比较无统计学意义以外,其他各组、各时间段内均抑制大鼠足趾肿胀。对棉球引起的大鼠肉芽肿实验中,JSTF低、中、高剂量组(125、250、500 mg/kg)与阿司匹林组(200 mg/kg)比较有明显的抑制作用,与模型组相比较有统计学差异(P0.05),其抑制率分别低剂量组为15.38%、中剂量组为18.48%、高剂量组为22.02%,阿司匹林组为43.11%。在蛋清和组胺引起大鼠足趾肿胀试验中,除了JSTF低剂量组(125mg/kg)在各个时间段内未能抑制大鼠足趾肿胀以外,其它给药组均明显的抑制了大鼠足趾肿胀,与模型组比较,有统计学意义(P0.05,P0.01)。(2)JSTF各剂量组能显著减少醋酸引起的小鼠扭体反应次数(P0.01)和延长小鼠热板痛阈值(P0.01,P0.05)。JSTF明显抑制第一和第二时相反应(P0.01,P0.05),而阳性对照组仅在第二时相出现抑制反应。(3)JFST可以提高免疫低下小鼠碳粒廓清指数K,尤其是500mg/kg剂量可使碳粒廓清指数显著升高(P0.01);与模型组比较JFST各剂量组也可以提高免疫低下小鼠血清溶血素生成(P0.05)。结论:JSTF具有明显的抗炎镇痛及免疫调节作用,其作用机制需要进一步研究。
[Abstract]:Aim: to study the anti-inflammatory, analgesic and immunomodulatory effects of JSTF in order to provide basic data for its further development. Methods the mice ear swelling induced by xylene, capillary permeability, cotton ball granuloma and egg white, histamine and carrageenin induced toe swelling in rats were used. Aspirin was used as the positive control. To observe the inhibitory effect of JSTF on inflammation in mice by acetic acid-induced writhing test, hot plate method and formaldehyde induced pain test. The analgesic effect of JSTF was observed. The effects of JSTF on nonspecific and specific humoral immune function in mice were observed by carbon particle clearance and serum hemolysin assay. Results JFST(125250500 mg / kg in low, middle and high dose groups could significantly inhibit the development of auricle swelling induced by xylene in mice (P 0.05). The inhibition rates were 40.67 and 51.78, respectively. JFST(125250500 mg / kg in low, middle and high dose groups also significantly inhibited the development of capillary permeability in mice (P 0.05), and the inhibition rates were 11.63% and 32.56%, respectively. In the experiment of carrageenin induced toe swelling in rats, except for the low dose group of JSTF and the model group in the first time period, there was no significant difference between the two groups, but in other groups, the swelling of the toes of rats was inhibited in each time period. In the experiment of cotton ball induced granuloma in rats, JSTF was significantly inhibited in the low, medium and high dose groups (125,250,500 mg / kg) and aspirin group (200mg / kg). Compared with the model group, the inhibition rate was 15.38 in the low dose group, 18.48 in the middle dose group, 22.02 in the high dose group and 43.11 in the aspirin group. In the test of toe swelling induced by egg white and histamine in rats, except for the low dose group of JSTF (125 mg / kg), which could not inhibit the swelling of rat toe in every time period, the other groups obviously inhibited the swelling of the toes of rats, compared with the model group. There was statistical significance that P0.05P0.01P0.01P0.05JSTF could significantly reduce the times of writhing reaction induced by acetic acid in mice and prolong the pain threshold of hot plate in mice P0.01P0.05N. JSTF significantly inhibited the first and 02:00 phase responses, while the positive control group only showed inhibition at 02:00. JFST could increase the clearance index of carbon particles in immunocompromised mice, especially the dosage of 500mg/kg could significantly increase the clearance index of P0.01G, and the serum hemolysin production of hypoimmunized mice was also increased by different doses of JFST compared with the model group. ConclusionJSTF has obvious anti-inflammatory, analgesic and immunomodulatory effects, and its mechanism needs further study.
