“百会”透“曲鬓”对急性期脑出血大鼠脑组织中AIF表达影响的实验研究

【摘要】:目的:通过针刺"百会"透"曲鬓",观察在针刺治疗过程中凋亡诱导因子AIF蛋白表达的变化,从而探讨其对急性期脑出血大鼠脑组织中AIF表达的影响,为临床上治疗急性期脑出血提供有利的依据。方法:75只雄性Wistar大鼠,随机分为空白组、模型组和针刺组。其中空白组15只大鼠、模型组和针刺组各30只,每组又平均分为6h,12h,24h,3d,7d五个时间点。模型组和针刺组采用自体血注入法建立脑出血模型。大鼠苏醒后观察并记录其神经功能缺损程度,挑选符合条件的模型,对针刺组进行针刺"百会"透"曲鬓"的治疗。随后对各组大鼠进行系统的实验观察:(1)观察各组大鼠神经功能缺损体征及评分情况;(2)采用TUNEL法检测各组大鼠脑出血后血肿周围细胞凋亡情况;(3)采用免疫组化法检测AIF在脑出血后血肿周围的动态表达。结果:(1)模型组和针刺组,在6h时开始出现神经功能缺损体征;12h-24h,神经功能缺损体征持续加重;在3d时,神经功能缺损体征最为严重,7d时神经功能缺损体征明显好转。针刺组神经功能评分较相同时间点模型组明显改善且在12、24h、3d时有显著差异(P0.01);(2)TUNEL法检测结果显示:与模型组比较,针刺组在各时间点神经细胞凋亡数量均出现不同程度的减少,且在24h、3d、7d时有显著差异(P0.01);(3)免疫组化检测结果显示:与模型组比较,针刺组在各时间点AIF蛋白表达水平均有不同程度的降低,且在24h、3d、7d时有显著差异(P0.01)。结论:(1)针刺"百会"透"曲鬓"可以减轻脑出血大鼠神经功能缺损体征;(2)针刺"百会"透"曲鬓"能够有效抑制脑出血后神经细胞凋亡的产生;(3)针刺"百会"透"曲鬓"可以减少凋亡诱导因子AIF的表达,通过抑制AIF的表达,对脑出血后血肿周围脑组织起到保护作用。
[Abstract]:Objective: to observe the expression of apoptosis-inducible factor (AIF) protein in the brain of rats with acute cerebral hemorrhage (ICH), and to investigate the effect of Acupuncture on the expression of Apoptosis-inducible Factor (AIF) in the brain of rats with acute intracerebral hemorrhage (ICH). To provide a favorable basis for clinical treatment of acute cerebral hemorrhage. Methods 75 male Wistar rats were randomly divided into blank group, model group and acupuncture group. There were 15 rats in the blank group, 30 rats in the model group and 30 in the acupuncture group. The model group and acupuncture group were used to establish the model of intracerebral hemorrhage by autologous blood injection. After waking up, the rats were observed and recorded the degree of nerve function defect, and the models were selected, and the acupuncture group was treated with acupuncture "Baihui" and "Baihui". Then the rats in each group were observed systematically. (1) the signs and scores of nerve function defect in each group were observed. (2) TUNEL method was used to detect apoptosis around hematoma in rats after intracerebral hemorrhage) Immunohistochemical method was used to detect the apoptosis of the cells around the hematoma. The dynamic expression of AIF around hematoma after intracerebral hemorrhage was measured. Results in the model group and the acupuncture group, the signs of nerve function defect began to appear at 6 h after 12 h 24 h, and the signs of nerve function defect continued to increase, and at 3 d, the signs of nerve function defect were obviously improved at 7 d after the most serious signs of nerve function defect. The score of nerve function in acupuncture group was significantly improved than that in model group at the same time point, and there was significant difference at 12 ~ 24 hours after 3 days. The results of Tunel method showed that the number of neuronal apoptosis in acupuncture group was decreased in varying degrees compared with model group. The results of immunohistochemical analysis showed that the expression of AIF protein in acupuncture group was significantly lower than that in model group at each time point, and there was a significant difference in P0.01protein expression at 24 h / 3 d ~ (-1) and 3 d / 7 ~ (th) after acupuncture compared with the model group, the expression of AIF protein in the acupuncture group was significantly lower than that in the model group at 7 days after the acupuncture treatment, and there was a significant difference between the two groups at 24 h / 3 d ~ (-1). Conclusion the acupuncture of "Baihui" ("Baihui") can reduce the signs of nerve function defect in rats with intracerebral hemorrhage. Acupuncture "Baihui" "diaphanous" can effectively inhibit the production of neuronal apoptosis after intracerebral hemorrhage and acupuncture "Baihui" can effectively inhibit the occurrence of neuronal apoptosis after intracerebral hemorrhage and acupuncture "Baihui" ("Baihui") can effectively inhibit the occurrence of neuronal apoptosis after intracerebral hemorrhage. The expression of apoptosis-inducing factor AIF could be reduced by "qu Si". By inhibiting the expression of AIF, it can protect the brain tissue around hematoma after intracerebral hemorrhage.
