不同时长艾灸对实验性KOA兔IL-1β、TNF-α影响的研究
[Abstract]:Objective:To observe the relationship between the content of IL-1 beta and TNF-a in the joint fluid and the expression of type II collagen protein and its mRNA in the articular cartilage of experimental KOA rabbits treated with Moxibustion of three different time lengths, and to explore the relationship between the effects of moxibustion of different time lengths on the treatment of KOA rabbits. Methods: Fifty Japanese white rabbits were randomly divided into blank group, model group, 10 min treatment group, 20 min treatment group and 40 min treatment group with 10 rabbits in each group. The experimental KOA model was duplicated by intracavitary injection of papain. The blank group was injected 0.5ml saline into the right knee. After modeling, the "inner knee eye" and "outer knee eye" acupoints of three moxibustion treatment groups were treated. The treatment group was treated for l0 minutes, the treatment group for 20 minutes and the treatment group for 40 minutes once a day, five times a course of treatment, with an interval of 2 days. The cartilage and synovium of the medial femoral condyle of the right knee joint and the adjacent tissues were taken out after 4 courses of treatment. The pathological morphology was analyzed by HE staining and Masson staining. The expression of type II collagen protein was detected by immunohistochemistry. The expression of type II collagen mRNA was detected by RT-PCR. The contents of IL-1beta and TNF-alpha in the joint fluid of the right knee joint were detected by Elisa method.The data obtained during the experiment were analyzed by SPSS21.0 and LSD was used for comparison among groups.The data were expressed by mean (+standard deviation) and P 0.05 respectively. Compared with the model group, the inflammatory cell infiltration in the model group was obvious; compared with the model group, the obvious inflammatory cell infiltration in the 10 minutes, 20 minutes and 40 minutes group was significantly reduced; compared with the 10 minutes, 20 minutes and 40 minutes group, the small amount of inflammatory cell infiltration in the 40 minutes group was significantly more than the other two groups; there was no significant difference between the l0 minutes group and 20 minutes group.2. Masson staining. OD value: Compared with the blank group, the OD value of the model group decreased significantly after Masson staining (p0.01); compared with the model group, the OD value of the 10 min, 20 min and 40 min group increased significantly (p0.05); compared with the 10 min, 20 min and 40 min group, the OD value of the 40 min group increased more significantly than the other two groups (p0.01); compared with the l0 min group, 20 min group had no statistical difference (p0.05). 3. Detection of IL-1beta and TNF-alpha in synovial fluid: Compared with the blank group, the levels of IL-1beta and TNF-alpha in the synovial fluid of the model group increased significantly (p0.01); Compared with the model group, the levels of IL-1beta and TNF-alpha in the synovial fluid of the 10 min, 20 min and 40 min groups decreased significantly (p0.01); Compared with the other two groups, the levels of IL-1beta and TNF-alpha in the 40 min group decreased significantly (p0.01). Detection of type II collagen protein expression and its mRNA expression level in articular cartilage: Compared with the blank group, the expression level of type II collagen protein and its mRNA in the model group was significantly lower (p0.01); compared with the model group, the expression level of type II collagen protein and its mRNA in the 10 min, 20 min and 40 min groups were significantly higher (p0.05). Compared with the other two groups, the expression of collagen type II protein and its mRNA in the 40-minute group were more significant (p0.05); there was no significant difference between the 20-minute group and the l0-minute group (p0.05). Conclusion: 1. The experimental KOA model could be successfully reproduced by intraarticular injection of papain into the knee joint of rabbits. Type 2. Inflammatory infiltration of synovium is an important factor in accelerating cartilage matrix decomposition and destroying articular cartilage during the pathological development of KOA. Moxibustion at "inner knee eye" and "outer knee eye" points for 3, 10, 20 and 40 minutes can control the inflammatory infiltration of synovium, increase the expression of type II collagen protein and decrease the content of IL-1beta and TNF-alpha in the joint fluid of experimental KOA rabbits. The expression level of mRNA delayed the degeneration of articular cartilage, the most significant in group 40min.
