电针刺激对大鼠受损脊髓节段HIF-1a、VEGF表达影响的研究

【摘要】:目的:研究电针刺激对脊髓损伤大鼠受损脊髓节段HIF-1a及VEGF表达的影响。方法:采用脊髓钳夹法复制雌性SD大鼠脊髓损伤模型,在造模成功第3天后,随机将大鼠分为夹脊组、胃经组和损伤对照组,分别运用电针刺激夹脊穴位、足阳明胃经穴位治疗以及不使用电针治疗。期间使用BBB评分法定期对各组大鼠神经功能评分进行评价,在治疗14天后处死大鼠,取各组大鼠受损脊髓节段脊髓,行HE染色观察组织受损情况;行免疫组织化学染色对比各组大鼠HIF-1a及VEGF蛋白表达情况;行qRT-PCR对比各组大鼠HIF-1a及VEGF的mRNA表达情况。结果:各组大鼠的BBB功能评分在治疗前,治疗1周均无明显显著差异,治疗2周时BBB评分:阳明组夹脊组损伤对照组;HE染色显示各组大鼠脊髓损伤情况符合其功能评分的表现;免疫组织化学染色结果显示,HIF-1a的表达在电针干预第7天:对照组阳明经组夹脊穴组,电针干预第14天:阳明经组夹脊穴组对照组,阳明经组表达量:第7天第14天,夹脊穴组表达量:第7天第14天,对照组表达量:第7天第14天,VEGF的表达情况在第7天:对照组阳明经组夹脊穴组,第14天:阳明经组夹脊穴组对照组,阳明经组表达量:第7天第14天,夹脊穴组表达量:第7天第14天,对照组表达量:第7天第14天;qRT-PCR结果显示,HIF-1a的表达在电针干预第7天:夹脊穴组对照组阳明经组,电针干预第14天:阳明经组夹脊穴组对照组,阳明经组表达量:第7天第14天,夹脊穴组表达量:第7天第14天,对照组表达量:第7天第14天,VEGF的表达情况在第7天:夹脊穴组对照组阳明经组,第14天:阳明经组夹脊穴组对照组,阳明经组表达量:第7天第14天,夹脊穴组表达量:第7天第14天,对照组表达量:第7天第14天。结论:电针可以通过提高受损脊髓节段HIF-1a及VEGF的蛋白和mRNA的表达来实现对SCI后功能障碍的恢复干预作用。电针刺激足阳明胃经对SCI后功能障碍治疗,最佳的介入时间为受损1周后。
[Abstract]:Aim: to study the effect of electroacupuncture on the expression of HIF-1a and VEGF in injured spinal cord in rats. Methods: the spinal cord injury model of female SD rats was established by spinal cord clamp method. On the 3rd day after the establishment of the model, the rats were randomly divided into three groups: Jiaji group, gastric meridian group and injury control group. Foot Yangming stomach meridian acupoint treatment and do not use electroacupuncture treatment. The neurological function of rats in each group was evaluated regularly by BBB scoring method. After 14 days of treatment, the rats were killed. The injured spinal cord of the rats in each group was taken out, and the damage of the spinal cord was observed by HE staining. The expression of HIF-1a and VEGF protein and the mRNA expression of HIF-1a and VEGF in each group were compared by immunohistochemical staining and qRT-PCR respectively. Results: before treatment, there was no significant difference in BBB functional score of rats in each group after 1 week of treatment. After 2 weeks of treatment, the BBB score: Yangming group with spinal cord injury control group, HE staining showed that the spinal cord injury in each group accorded with its functional score. Immunohistochemical staining showed that the expression of HIF-1a was observed on the 7th day of electroacupuncture intervention: the control group was treated with Yangming Meridian Jiaji acupoint group, the control group with electroacupuncture intervention on the 14th day, the Yangming Meridian group with Jiaji acupoint group, and the Yangming Meridian group with the expression level of Yangming Meridian Group on the 7th day on the 14th day. The expression of VEGF in Jiaji acupoint group on the 7th day and the control group on the 14th day: on the 7th day, the expression of VEGF was on the 7th day; in the control group, the Yangming meridian group was the Jiaji acupoint group, the 14th day was the Yangming meridian Jiaji acupoint group, the control group was the Yangming meridian Jiaji acupoint group. The expression of Yangming Meridian on the 14th day, Jiaji acupoint group on the 14th day, the control group on the 14th day and on the 14th day on the 7th day. QRT-PCR results showed that the expression of HIF-1a was on the 7th day after electroacupuncture intervention: the Jiaji acupoint group was the control group, and the control group was Yangming meridian group. The 14th day of electroacupuncture intervention: Yangming meridian group Jiaji point group control group, Yangming meridian group expression amount: the 7th day 14 days, Jiaji point group expression amount: the 7th day the 14th day; The expression of VEGF in the control group: on the 7th day, the expression of VEGF was on the 7th day: Jiaji acupoint group, the control group, the Yangming meridian group, the 14th day: the Yangming meridian group, the control group, and the Yangming meridian group on the 7th day and the 14th day, respectively. The expression level of Jiaji acupoint group was 14 days on the 7th day, and that of the control group was 14 days on the 7th day. Conclusion: electroacupuncture can improve the expression of HIF-1a and VEGF protein and mRNA in injured spinal cord segment to improve the recovery of SCI. The best interventional time was 1 week after injury after SCI was treated by electroacupuncture stimulation of foot Yangming stomach Meridian.
