大鼠脑外伤后BDNF的表达及其死后稳定性的研究

【摘要】:目的:探讨脑外伤后脑源性神经营养因子(Brain derived neurotrophical factor,BDNF)表达的变化和损伤时间的关系及其死后稳定性。方法:将健康SD大鼠105只,随机分成4组:正常组、假手术组、脑外伤组、死后稳定性组。采用重物砸伤脑组织建立脑外伤模型(创伤组、稳定性组),运用免疫组化和图像分析技术,检测各组BDNF的表达变化规律。结果:与正常组、假手术组比较,损伤后0.5 h,BDNF的免疫反应阳性细胞在损伤区周围增多,损伤后6 h BDNF阳性细胞数达高峰,以后逐渐下降,24 h后降至接近正常;死后稳定性组各时间点BDNF表达无显著性差异(P0.05)。结论:脑外伤后BDNF表达与损伤时间具有相关性;在-20℃条件下,死后稳定性好。
[Abstract]:Aim: to investigate the relationship between the expression of brain-derived neurotrophic factor (BDNF) and the time of injury and postmortem stability after traumatic brain injury. Methods: 105 healthy SD rats were randomly divided into 4 groups: normal group, sham operation group, brain injury group and postmortem stability group. The brain injury model (trauma group, stability group) was established by heavy object injury. The expression of BDNF in each group was detected by immunohistochemistry and image analysis. Results: compared with normal group and sham operation group, the number of BDNF immunoreactive cells increased around the injured area at 0.5 h after injury, reached the peak at 6 h after injury, then decreased to almost normal at 24 h after injury. There was no significant difference in BDNF expression at all time points in postmortem stability group (P0.05). Conclusion: the expression of BDNF is correlated with the time of injury, and the postmortem stability is good at-20 鈩,
