漳平市杜鹃花产业调查与VIGS技术体系初步构建

【摘要】:本研究通过文献检索、实地走访和电话咨询等方式进行漳平市杜鹃花产业现状分析,从中发现杜鹃产业存在的问题,最后提出相应对策,旨在促进漳平市杜鹃花产业健康持续发展。目前漳平市杜鹃花产业存在品种引种为主,缺乏自主培育品种问题,同时杜鹃花多以杂交育种为主,所需时间长,品种更新缓慢,而VIGS技术作为一种简单、快速、高通量的基因功能分析方法,还未应用到杜鹃中,因此本研究开展了杜鹃花VIGS技术的体系构建,进行杜鹃花观赏性相关基因的功能验证,利于漳平市杜鹃花新品种的培育,从而促进杜鹃产业发展。本文的主要研究结果如下:1.对漳平市杜鹃花产业的生产情况、商品情况、发展优势和经营情况进行调查,结果如下:(1)龙岩作为福建省杜鹃花产业的主产区,2015年种植面积达400 hm2,占福建省87%;(2)漳平市杜鹃花产业规模虽然有所缩小,甚至在2013年出现产值下滑18%的情况,但是近两年漳平市专业化生产水平逐步成熟、生产技术不断提高,经济效益不断提高,产值呈现上升趋势(3)搜集到的漳平市杜鹃花产业中约100个杜鹃花品种,从中发现的杜鹃商品花色主要为红色和紫色,花形特征为大花和重瓣;盆花和苗木为主要的商品类型,盆景杜鹃是新兴的商品类型(4)漳平市杜鹃花产业由于土地与租金问题,主要以个体农户为经营模式;销售模式大部分为传统的批发,开店,进驻商场的方式,互联销售模式逐渐兴起。2.漳平市杜鹃花产业发展中存在的问题有:(1)商品化品种依赖引种,缺乏自主知识产权品种;(2)产品类型和产品应用创新能力不足;(3)产业运营有待完善,花企缺乏综合竞争力。3.漳平市杜鹃花产业发展的优势条包括:适宜的气候环境和独特的区位交通优势,悠久的杜鹃花栽培历史,政府部门和相关政策的支持,丰富的种质资源,充沛的劳动力,以及良好的花卉市场发展潜力。4.通过对漳平市杜鹃花产业存在的问题和其优势条件的分析,提出了一系列有助于漳平市杜鹃花产业发展的对策:(1)重视新品种研发,提升品种创新能力;(2)丰富产品类型,拓宽产品应用;(3)加强产学研合作,提升花企综合竞争力;(4)提升产业现代化水平,优化产业运营。5.基于烟草脆裂病毒载体,进行杜鹃VIGS病毒载体的构建。成功克隆了杜鹃392 bp的PDS基因片段、472bp的CHS基因片段、400bp的DFR基因片段、372bp的RDR2A基因片段和350bp的MADS基因片段。同时构建了杜鹃VIGS病毒载体包括,VIGS-PDS 载体、VIGS-CHS 载体、VIGS-DFR 载体、VIGS-MADS载体、VIGS-RDR2A载体。6.杜鹃VIGS技术体系的建立与优化。以杜鹃嫩叶和成熟叶为实验材料,设置不同处理的侵染液浓度OD600值分别为1.0、1.5和2.0,采用注射法将含有VIGS-PDS载体的农杆菌瞬时转化杜鹃植株。通过对侵染后叶片的表型观察和统计,发现杜鹃嫩叶和成熟叶均出现PDS基因表达受抑制的表型。初步表明采用pTRV病毒载体可以用于杜鹃植株的基因沉默。并且在综合分析了不同侵染液浓度(OD600值)对杜鹃叶片PDS基因沉默的效果,以及对叶片生长发育的影响,确定了较适宜的侵染液OD600值为1.0。7.杜鹃花器官VIGS技术的构建与优化。设置OD600值为1.0和2.0两种侵染液浓度,采用注射法和涂抹法处理杜鹃的花苞和半开放的花苞的基部,以及开放花的花瓣。对侵染后的花器官进行表型观察和统计,发现开放花均未出现预期的CHS基因、DFR基因、MADS基因和RDR2基因表达受抑制的表型。但通过比较分析各组实验处理结果,本文对杜鹃花器官VIGS体系的构建提出了一些值得参考的建议。
[Abstract]:In this study, the current situation of Rhododendron industry in Zhangping was analyzed through literature search, field visit and telephone consultation, and the existing problems of Rhododendron industry were found. Finally, the corresponding countermeasures were put forward to promote the healthy and sustained development of the azalea industry in Zhangping. At present, the azalea industry in Zhangping was mainly introduced and lack of independent culture. At the same time, the Rhododendron is mostly hybrid breeding, with long time and slow variety, and VIGS technology is not used in Rhododendron as a simple, fast, high throughput gene function analysis method. Therefore, the system of Rhododendron VIGS is constructed and the function of Rhododendron ornamental related genes is tested. It is beneficial to the cultivation of new Rhododendron varieties in Zhangping to promote the development of Rhododendron industry. The main research results of this paper are as follows: 1. the production situation, commodity situation, development advantage and management situation of the azalea industry in Zhangping are investigated. The results are as follows: (1) Longyan is the main producing area of the Rhododendron industry in Fujian Province, and the planting area is 40 in 2015. 0 Hm2, accounting for 87% in Fujian province; (2) although the size of the Rhododendron industry in Zhangping has narrowed, even in 2013, the output value of the city has declined by 18%. But in the last two years, the professional production level of Zhangping is gradually mature, the production technology is increasing, the economic benefit is increasing, and the output value is rising (3) of the Rhododendron industry of Zhangping, which is about 1. 00 Rhododendron varieties, from which Rhododendron is mainly red and purple, flower shape features large flowers and heavy petals, potted flowers and seedlings are the main commodity types, and bonsai Rhododendron is a new commodity type (4) the Rhododendron industry in Zhangping city is mainly based on individual farmers as the management model because of land and rent. The existing problems in the development of.2. Zhangping azalea industry are as follows: (1) commodity variety dependence introduction, lack of independent intellectual property rights variety; (2) product type and product application innovation ability is insufficient; (3) industry operation needs to be improved, flower enterprises lack comprehensive competitiveness. .3. Zhangping Rhododendron industry development advantages include: suitable climate environment and unique regional traffic advantages, long history of Rhododendron cultivation, the support of government departments and related policies, rich germplasm resources, abundant labor force, and good flower market development potential.4. through the existence of the Rhododendron industry in the city of Zhangping The problem and the analysis of its predominance, put forward a series of countermeasures to help the development of Zhangping Rhododendron industry: (1) attach importance to new varieties research and development, improve the ability of variety innovation; (2) enrich product types, broaden the application of products; (3) strengthen the cooperation of production, research and research, improve the comprehensive competitiveness of the enterprises; (4) upgrade the level of industrial modernization and optimize the industrial operation.5. Based on the tobacco embrittlement virus vector, the vector of Rhododendron VIGS virus was constructed. The PDS gene fragment of Rhododendron 392 BP, the CHS gene fragment of 472bp, the DFR fragment of 400bp, the RDR2A gene fragment of 372bp and the MADS gene fragment of 350bp were successfully cloned. The establishment and optimization of R carrier, VIGS-MADS carrier, VIGS-RDR2A vector.6. Rhododendron VIGS technology system, with Rhododendron leaves and mature leaves as experimental materials, the OD600 values of different treatments were 1.0,1.5 and 2 respectively, and the Agrobacterium containing VIGS-PDS carrier was transformed into Rhododendron by injection method. Phenotypic observation and statistics showed that PDS gene expression was inhibited in both young and mature leaves of Rhododendron. It was preliminarily indicated that the use of pTRV virus vector could be used in the gene silencing of Rhododendron plants, and the effect of PDS gene silencing on the leaves of Rhododendron (OD600) and the growth and development of leaves were analyzed in a comprehensive analysis. The optimum OD600 value of the OD600 was to construct and optimize the VIGS technology of the 1.0.7. Rhododendron organ. The concentration of OD600 was 1 and 2, the base of the bud and the semi open bud of Rhododendron and the open flower petals were treated with the injection method and the smear method. The phenotypic observation and statistics of the flower organs after the infection were observed. It was found that the expected CHS gene, the DFR gene, the MADS gene and the RDR2 gene were inhibited in the open flower, but by comparing and analyzing the results of the experimental treatment, some suggestions for the construction of the VIGS system of the azalea organ were put forward.
