心肌细胞分离与培养的教学内容与方法探讨

【摘要】:培养心肌细胞是生物医学研究领域一项新实验技术,在心血管病理生理学研究中起着相当重要的作用。本学科自1998年为研究生开设《心肌细胞分离与培养》选修课,目的是使学生掌握心肌细胞分离培养的基本原理和方法并应用于科学研究中。讲义编写和讲授内容以原代细胞培养为线索,以大鼠心肌细胞分离和培养为实例,比较详尽地讲解和演示了实验的全过程。主要内容:(1)分离:①未成熟心肌细胞:准备→取材→酶解→离心→制备细胞悬液;②成熟心肌细胞:准备Langendroff装置→摘取心脏和插管→灌流→酶解→离心复钙。(2)培养:包括贴壁培养和悬浮培养。重点讲授纯化心肌细胞、酶选择、消化程度把握、各部位心肌细胞培养、2种培养方法差异与表达的特性、生长状态等。(3)建立相关疾病模型:如心肌肥厚、心衰等。(4)应用:包括心肌细胞超微结构、细胞内外离子转运通道、细胞凋亡、受体、蛋白质组学、药物学研究等。该方法稍加改良亦可用于鸡、小鼠、兔、狗等动物及人类胎儿和成人手术获得的心肌碎片的心肌细胞培养。通过多年探索,已基本完善了教学内容,提高了教学质量和教学水平。
[Abstract]:Cardiomyocyte culture is a new experimental technique in biomedical research, which plays an important role in cardiovascular pathophysiology. The course of cardiomyocyte separation and culture has been offered to graduate students since 1998. The purpose of this course is to make students master the basic principles and methods of cardiomyocyte separation and culture and apply them to scientific research. Taking primary cell culture as clue and rat cardiomyocyte isolation and culture as an example, the whole process of the experiment is explained and demonstrated in detail. The main contents were as follows: (1) isolation: 1immature cardiomyocytes: preparation of cell suspensions by centrifugation; (2) mature cardiomyocytes: preparation of Langendroff device for the extraction of cardiac cells and the centrifugation of pericardial pericardium. (2) culture, including adherent culture and suspension culture. Emphasis is placed on purified cardiomyocytes, enzyme selection, digestion degree assurance, cardiomyocyte culture in different parts, differences and expression characteristics of two culture methods, growth state, etc. (3) establishing models of related diseases, such as myocardial hypertrophy, and so on. (4) applications include ultrastructure of cardiomyocytes, intracellular and extracellular ion transport channels, apoptosis, receptors, proteomics, pharmacology, and so on. The method can also be used to culture cardiac muscle cells from chickens, mice, rabbits, dogs, human fetuses and adults. Through many years of exploration, the teaching content has been basically improved, and the teaching quality and teaching level have been improved.
