荷叶黄酮营养品配方的减重作用及其机制的实验研究

【摘要】:目的:根据对运动员减控体重的要求和特点,选择设计荷叶黄酮配方,并进行本营养品的减重降脂作用及安全性实验研究。方法:采用体外酶抑制法测定荷叶黄酮对脂肪酶活性的影响;采用营养性肥胖大鼠模型试验进行荷叶黄酮的减重和降血脂作用研究。MTT法检测不同浓度荷叶黄酮对3T3-L1前脂肪细胞增殖的影响。油红染色法定量检测荷叶黄酮对3T3-L1前脂肪细胞分化的影响。结果:荷叶黄酮对脂肪酶活性有抑制作用,且随着浓度的增大而增强。肥胖大鼠模型试验结果表明,荷叶黄酮能显著降低动物的体重,体脂比,血清甘油三酯(TG),胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)以及HMG-Co A还原酶活性。MTT法检测结果显示,荷叶黄酮可抑制3T3-L1前脂肪细胞的分化,其抑制作用具有浓度依赖性。结论:补充荷叶黄酮配方营养品具有明显的减重作用,其作用机制与降低血脂、抑制脂肪细胞分化有关。
[Abstract]:Objective: according to the requirements and characteristics of weight reduction and control for athletes, the formula of lotus leaf flavonoids was selected and the effects and safety of the nutrient on reducing body weight and lipid were studied. Methods: the effect of flavone on lipase activity in lotus leaves was determined by enzyme inhibition method in vitro. The effects of flavonoids from lotus leaves on the proliferation of preadipocytes before 3T3-L1 were studied by MTT assay. The effects of flavonoids from lotus leaves on the differentiation of preadipocytes from 3T3-L1 were detected quantitatively by oil red staining. Results: flavonoids of lotus leaf inhibited lipase activity and increased with the increase of concentration. The results of obese rat model showed that flavonoids from lotus leaves could significantly reduce the body weight, body fat ratio, serum triglyceride, cholesterol triglyceride, low density lipoprotein cholesterol (LDL-C) and HMG-Co A reductase activity. Lotus leaf flavonoids could inhibit the differentiation of 3T3-L1 preadipocytes in a concentration dependent manner. Conclusion: supplementation of flavonoids from lotus leaves has obvious weight loss effect, and its mechanism is related to lowering blood lipid and inhibiting adipocyte differentiation.
