人宫颈癌Hela细胞TPX2基因沉默对放射敏感性的影响

【摘要】:目的观察TPX2基因沉默的人宫颈癌Hela细胞对X射线的敏感性。方法将宫颈癌Hela细胞分为TPX2-siRNA组、NC-siRNA组、Control组,TPX2-siRNA组转染TPX2-siRNA,NC-siRNA组转染NC-siRNA即空载体siRNA,Control组仅加入转染试剂。采用实时荧光定量PCR、Western blot分别检测各组细胞中的TPX2 mRNA与蛋白。采用CCK8法检测不同剂量(0、2、4、6 Gy)X线照射后TPX2-siRNA组细胞的增殖抑制率。采用流式细胞术检测4 Gy X线照射前后TPX2-siRNA组细胞的凋亡率。结果与NC-siRNA组、Control组相比,TPX2-siRNA组细胞TPX2 mRNA、蛋白相对表达量降低(P均0.05)。TPX2-siRNA组细胞接受0、2、4、6 Gy X线照射后,细胞增殖抑制率分别为4.71%±0.15%、5.26%±0.69%、6.64%±0.13%、15.19%±0.73%,两两组间比较P均0.05。TPX2-siRNA组细胞接受4 Gy X线照射前后细胞凋亡率分别为11.43%±0.03%、12.68%±0.03%,两者比较P0.05。结论 TPX2基因沉默的人宫颈癌Hela细胞对X射线的敏感性增加。
[Abstract]:Objective to observe the radiosensitivity of human cervical cancer Hela cells silenced with TPX2 gene to X-ray. Methods Cervical cancer Hela cells were divided into TPX2-siRNA group and NC-siRNA group. Control group was transfected with TPX2-siRNA group and NC-siRNA group was transfected into NC-siRNA vector siRNA. In Control group, only transfection reagent was added and real-time quantitative PCR was used. Western blot was used to detect the TPX2 mRNA and protein in the cells of each group, and different doses of TPX2 mRNA and protein were detected by CCK8 method. 6 Gy). The proliferation inhibition rate of TPX2-siRNA group after X-ray irradiation was determined by flow cytometry. The apoptosis rate of TPX2-siRNA group before and after X-ray irradiation was compared with that of NC-siRNA group. Compared with the TPX2 mRNAs of the Control group, the relative protein expression of the Control group was lower than that of the TPX2-siRNA group. The cells in the TPX2-siRNA group received 0. 2After X ray irradiation, the inhibition rate of cell proliferation was 4.71% 卤0.15 and 5.26% 卤0.69 and 6.64% 卤0.13%, respectively. 15.19% 卤0.73%. The apoptosis rate of the cells in the TPX2-siRNA group was 11.43% 卤0.03% before and after 4 Gy X-ray irradiation. Conclusion TPX2 gene silencing human cervical cancer Hela cells is more sensitive to X-ray than P0.05.Conclusion TPX2 gene silencing human cervical cancer Hela cells is more sensitive to X ray.
