小干扰RNA沉默YKL-40基因对子宫内膜癌HEC-1A细胞顺铂化疗敏感度的影响

【摘要】:目的通过si RNA沉默YKL-40基因探讨其对子宫内膜癌HEC-1A细胞顺铂化疗敏感度的影响。方法 si RNA基因干扰技术沉默YKL-40基因,将人子宫内膜癌HEC-1A细胞分为三组:空白对照组、空载体组、si RNA实验组。q RT-PCR法检测沉默效率。q RT-PCR法检测顺铂处理子宫内膜癌细胞对癌细胞中YKL-40基因的影响。MTT法和流式细胞术检测靶细胞的增殖能力以及凋亡率。结果转染组YKL-40m RNA的表达量显著低于空白对照组和空载体组(P=0.036,P=0.005),经过相同浓度的顺铂处理子宫内膜癌后,YKL-40基因的表达量上调。转染组的增殖能力低于空白对照组和空载体组(P0.05),而凋亡率高于空白对照组和空载体组(P=0.000,P=0.000)。结论 YKL-40基因沉默可以提高子宫内膜癌HCE-1A细胞对顺铂的敏感度。YKL-40基因具有抗凋亡作用。
[Abstract]:Objective to investigate the effect of si RNA silencing YKL-40 gene on chemotherapeutic sensitivity of HEC-1A cells with endometrial carcinoma. RNA gene interference technique silenced YKL-40 gene. Human endometrial carcinoma HEC-1A cells were divided into three groups: blank control group and empty vector group. Si RNA experimental group. Q RT-PCR method to detect silencing efficiency. Q. RT-PCR assay to detect the effect of cisplatin treatment on YKL-40 gene in endometrial cancer cells. MTT assay and flow cytometry were used to detect the proliferation ability and apoptosis rate of target cells. L-40m. The expression of RNA was significantly lower than that of blank control group and empty vector group (. P0. 036. The expression of YKL-40 gene in endometrial carcinoma treated with the same concentration of cisplatin was up-regulated. The proliferation ability of transfected group was lower than that of blank control group and empty vector group (P0.05). The rate of apoptosis was higher than that of control group and empty carrier group. Conclusion YKL-40 gene silencing can increase the sensitivity of HCE-1A cells to cisplatin. YKL-40 gene has anti-apoptosis effect.
