DHPLC法分析视黄醇结合蛋白4基因rs17484721及rs36035572多态性与老年人超重肥胖的关系

【摘要】:目的利用聚合酶链式反应(PCR)-变性高效液相色谱(DHPLC)法对老年人群视黄醇结合蛋白(RBP)4基因的rs17484721和rs36035572位点进行筛查,并分析各基因型之间肥胖、胰岛素及血脂的差异。方法选择健康查体的汉族老年人304例。检测空腹血糖(FPG)、空腹胰岛素(FINS),计算胰岛素抵抗指数(HOMA-IR)。采用DHPLC技术筛查RBP4基因rs17484721和rs36035572多态性位点,测序分析结果。结果两个位点的突变存在强连锁不平衡。TT-II组体重指数(BMI)、腰臀比明显高于TC-ID+CC-DD组,考虑TT-II基因型的人群出现向心性肥胖的概率较大。结论 RBP4基因多态性可能与老年人超重肥胖相关。
[Abstract]:Objective to screen the rs17484721 and rs36035572 loci of retinol binding protein (RBP4) gene in the elderly by polymerase chain reaction (PCR) and denaturing high performance liquid chromatography (HPLC), and to analyze the obesity among different genotypes. Methods the difference of insulin and serum lipids was studied in 304 healthy subjects of Han nationality. Fasting blood glucose (FPG), fasting insulin (Fin) and insulin resistance index (HOMA-IRN) were measured. DHPLC technique was used to screen RBP4 gene rs17484721 and rs36035572 polymorphisms. Results there was strong linkage disequilibrium between the two loci. The body mass index (BMI) of TT-II group was significantly higher than that of TC-ID CC-DD group, and the WHR was significantly higher in TT-II group than in TC-ID CC-DD group. ConclusionThe RBP4 gene polymorphism may be associated with overweight and obesity in the elderly.
