绵羊ZBED6基因的遗传变异及其与胴体性状的相关性研究

【摘要】:为明确绵羊ZBED6基因的遗传变异,探讨ZBED6基因的遗传变异与绵羊主要胴体性状的相关性,本论文选择了新疆地方品种阿勒泰羊与特克赛尔羊的杂交后代(横交F2代、回交F2代2个群体)的225个个体为主要研究对象,以绵羊ZBED6基因为候选基因,旨在查明ZBED6基因的分子遗传特征及其在群体间的差异,分析该基因变异对所选绵羊胴体性状的影响,评价其作为所选绵羊重要生产性状分子遗传标记的利用价值。本研究的主要研究内容和结果如下:1.试验群体胴体性状的测定和影响因素分析试验测定了170只横交F2代群体和55只回交F2代群体的7个主要的胴体性状,包括胴体重量、背部脂肪厚度、眼肌面积、尾脂重量、胴体长、背部肌肉厚、背最长肌质量。比较了不同杂交方式对于胴体性状的影响发现横交F2群体的眼肌面积显著大于回交F2代群体(P0.05),横交F2代背部肌肉厚度极显著大于回交F2代群体(P0.01),横交F2代背最长肌重量极显著大于回交F2代(P0.01),横交F2代尾脂重量极显著小于回交F2代群体(P0.01),横交F2代胴体长比回交F2代群体长约1.65cm,接近于显著水平(P=0.05),两个杂交群体中的背部脂肪厚度和胴体重量差异不显著(P0.05);比较了不同性别对胴体性状的影响,发现公羊的眼肌面积显著大于母羊(P0.05),公羊的背部脂肪厚度极显著大于母羊(P0.01),公羊尾脂重量较母羊尾脂重2.87kg,接近于显著水平(P=0.05),母羊的胴体长极显著长于公羊(P0.01),不同性别的胴体重量,背部肌肉厚度和背最长肌重量差异不显著(P0.05)。2.绵羊ZBED6基因多态性检测针对ZBED6基因的全基因组长为3099bp的基因序列,本研究设计了5对PCR引物、样本量为225个个体的DNA进行扩增,经对结果分析后发现在Z-2引物测序中两个杂交群体均存在多态性,在C1723T和A2095G碱基处发生了碱基突变CT和AG。这两个突变均为同义突变。经卡方检验分析,基因型和等位基因分布在2个杂交群体中差异不显著(P0.05)。由于Z-1、Z-3、Z-4、Z-5两对引物经检测分析得到的突变频率较低,该四对引物均不宜进行下一步实验。3.ZBED6基因多态性与7个主要胴体性状的相关性分析通过对225个个体C1723T和A2095G两个位点的基因型检测ZBED6基因多态性信息,根据实验1的结果,考虑群体和性别影响因素,利用一般线性模型对7个胴体性状与ZBED6基因多态性进行关联分析。统计分析表明,这2个位点与7个胴体性状无关联性。ZBED6基因这2个突变位点是否与其他胴体性状相关,需要扩大样本数量并且与其余胴体表型进行关联分析验证。本论文结论:试验选取新疆地方品种阿勒泰羊与特克赛尔羊杂交后代的225个个体的耳组织样品进行ZBED6基因SNPs检测,并与该群体屠宰获得的7个胴体性状进行关联分析。研究表明,ZBED6基因编码区在受试群体中存在多态性,在C1723T和A2095G碱基处发生了碱基突变CT和AG,这两个位点的基因型与胴体性状不相关。同时,本研究对供试绵羊群体间及其性别对胴体性状的影响的分析显示,群体间眼肌面积差异显著,尾脂重、背部肌肉厚、背最长肌重差异极显著。其中,横交F2代羊群体在眼肌面积、背最长肌重、背部肌肉厚性状方面优于回交F2代群体,回交F2代群体尾脂重明显高于横交代羊群体;不同性别间眼肌面积、胴体长、背部脂肪厚度差异极显著。公羊眼肌面积优于母羊,但其背部脂肪厚度大于母羊。由此可见,横交F2代羊能够满足绵羊产肉相关性状的选育。
[Abstract]:For the clear genetic variation of ZBED6 gene in sheep, to investigate the relationship between genetic variation and carcass traits of main sheep ZBED6 gene, this paper chose the hybrid offspring of Xinjiang local varieties of Aletai sheep and Texel Sheep (cross backcross F2 generation, F2 generation 2 groups) and 225 individuals as the main object of study, to sheep ZBED6 gene as a candidate gene, aimed at identifying the molecular genetic features of ZBED6 gene and the difference between groups, analysis of the impact of the genetic variation of selected sheep carcass traits, evaluate the utilization value of the selected sheep as important production traits of molecular genetic markers. The main research contents and results are as follows: the determination of 7 major and the 170 cross carcass traits in F2 generation and 55 backcross F2 populations were measured in 1. test groups analysis of factors affecting carcass traits, including carcass weight, backfat thickness, eye Muscle area, tail fat weight, carcass length, back muscle thickness and longissimus muscle quality. Compare the different ways of hybridization influence on carcass traits that cross F2 groups were significantly greater than the eye muscle area (P0.05) of backcross F2 generation, F2 generation cross back muscle thickness was significantly higher than that of F2 group (back P0.01), cross F2 longissimus muscle weight was significantly higher than that of backcross F2 generation (P0.01), cross F2 tail fat weight was significantly lower than that of backcross F2 generation (P0.01), cross F2 generation backcross F2 generation carcass length ratio is about 1.65cm, close to significant level (P=0.05), two a hybrid group back fat thickness and carcass weight did not differ significantly (P0.05); the influence of different gender on carcass traits were compared, found that the ram of the eye muscle area was significantly greater than ewes (P0.05), the back fat thickness of rams was significantly higher than that of ewes (P0.01), sheep tail fat weight than sheep tail fat weight 2.87kg That is close to significant level (P=0.05), ewe carcass length was significantly longer than the RAM (P0.01), carcass weight of different sex, back muscle thickness and longissimus muscle weight had no significant difference (P0.05) of.2. sheep ZBED6 gene polymorphism detection of whole genome length for ZBED6 gene sequence of 3099bp genes, this research 5 pairs of PCR primers were designed, samples were amplified for 225 individuals of DNA, through the analysis of the results found in the Z-2 two hybrid population sequencing primers were found polymorphic, the mutation of CT and AG. of the two mutations were synonymous mutations in C1723T and A2095G bases. By chi square test analysis of genotype and allele distribution in 2 hybrid groups had no significant difference (P0.05). The Z-1, Z-3, Z-4, Z-5, two pairs of primers were detected by analysis of the mutation frequency is low, the four pairs of primers of.3.ZBED6 gene were not suitable for the next experiment Correlation analysis of normality and the 7 main carcass traits by the genotypes of 225 individuals of C1723T and A2095G two loci of ZBED6 gene polymorphism detection information, according to the results of the 1 experiment groups and gender influence factors, analyzing the correlation between the 7 carcass traits and ZBED6 gene polymorphism by using general linear model statistics. Analysis shows that these 2 loci and 7 carcass traits were unrelated.ZBED6 genes of the 2 mutations are associated with other carcass traits, need to expand the sample number and correlation analysis verification with the rest of the carcass phenotype. Conclusion: the experiment selects Xinjiang local varieties of Aletai sheep and Texel Sheep hybrids 225 individual ear tissue samples of ZBED6 SNPs gene detection, and 7 carcass traits were obtained with the group slaughter for correlation analysis. The results show that the ZBED6 gene encoding region in the tested group The polymorphism of the body, the mutation of CT and AG in C1723T and A2095G at base, not related to genotype and carcass traits of these two sites. At the same time, the research and Analysis on the effects of the tested sheep populations and sex on carcass traits showed significant difference between groups of eye muscle area, fat tail weight, back muscle thickness, the difference of longissimus muscle weight significantly. Among them, cross the F2 generation of sheep populations in the loin eye area, longissimus muscle weight, back muscle thickness traits than backcross progeny F2, backcross F2 generation tail fat weight was significantly higher than that of sheep cross account groups; gender differences and eye muscle area carcass length, back fat thickness, the difference was very significant. The eye muscle area is better than that of ram ewe, but the back fat thickness greater than ewes. Thus, cross the F2 generation of sheep meat producing sheep breeding can meet related traits.
