加味牛蒡子汤促进TNF-α转基因小鼠淋巴管回流功能防治类风湿关节炎的实验研究

【摘要】:目的观察加味牛蒡子汤(JWNBZT)对类风湿关节炎(RA)模型动物TNF-α转基因(TNF-Tg)小鼠关节炎症和关节周围淋巴回流功能的影响。方法将16只清洁级3月龄左右TNF-Tg小鼠按照体重随机分为TNF-Tg组和JWNBZT组,每组8只。另取10只同窝对照体重相似的野生型小鼠(WT)作为对照组,JWNBZT组每天以JWNBZT灌胃,对照组和TNF-Tg组每天以等量纯净水灌胃,治疗12周后小鼠足底皮下注射吲哚菁绿(ICG)测量小鼠淋巴管回流功能,并取踝关节制作石蜡切片进行苏木素-伊红(HE)染色观察炎性浸润和关节骨质破坏情况,抗酒石酸酸性磷酸酶(TRAP)染色观察破骨细胞数量。结果与对照组比较,TNF-Tg组小鼠淋巴管回流功能出现明显损害,Clearance和Pulse明显下降,淋巴管渗漏面积增多,踝关节内出现明显炎症浸润且骨质减少,距骨内破骨细胞增多(P0.05);与TNF-Tg组比较,JWNBZT组小鼠淋巴管回流功能明显增强,Clearance和Pulse升高,淋巴管渗漏面积减少,踝关节内炎症浸润减轻且骨质增多,距骨内破骨细胞减少(P0.05)。结论 JWNBZT能够减轻RA受累关节的炎症浸润和关节骨质破坏,并促进淋巴管回流功能,这可能是其治疗RA的机制。
[Abstract]:Objective to observe the effect of JWNBZT (JWNBZT) on arthritis and peripheral lymphoid reflux in TNF- 伪 transgenic TNF-Tg mice. Methods Sixteen TNF-Tg mice of 3 months old of clean grade were selected to follow their body weight according to their body weight. The machine was divided into TNF-Tg group and JWNBZT group. Eight mice in each group were taken as control group, and 10 wild type mice with similar body weight were taken as control group. JWNBZT group was treated with JWNBZT daily, and the control group and TNF-Tg group were treated with the same amount of pure water daily. After 12 weeks of treatment, the subcutaneous injection of indocyanine green (ICG) into the plantar of mice was used to measure the function of lymphatic reflux in mice, and paraffin sections were taken from the ankle joint to be stained with hematoxylin and eosin (HEH) to observe the inflammatory infiltration and joint bone destruction. The number of osteoclasts was observed by tartrate-resistant acid phosphatase (TRAP) staining. Results compared with the control group, the lymphatic reflux function of TNF-Tg group decreased significantly, and the lymphatic vessel leakage area increased. Significant inflammatory infiltration and osteopenia were observed in the ankle joint, and osteoclasts increased in the talus (P 0.05). Compared with the TNF-Tg group, the lymphatic reflux function of the JWNBZT group was significantly increased, and the lymphatic vessel leakage area was decreased in the JWNBZT group, and the function of lymphatic reflux was significantly increased in the JWNBZT group. Conclusion JWNBZT can reduce inflammatory infiltration and joint bone destruction and promote lymphatic reflux function, which may be the mechanism of JWNBZT in the treatment of RA.
