PIWI蛋白家族基因在绵羊生殖腺中的表达研究

【摘要】：为了研究PIWI基因对绵羊生殖细胞发育的影响,试验采用同源基因克隆的方法,以绵羊睾丸、卵巢RNA为模板,通过PCR扩增,半定量PIWI基因家族在苏尼特羊、小尾寒羊生殖腺的表达。结果表明:公羊PIWI蛋白家族3个成员mRNA的相对表达水平均极显著高于母羊(P0.01),PIWI1、PIWI2、PIWI4在高繁小尾寒羊母羊、低繁苏尼特母羊均只有微弱表达,同性别高繁小尾寒羊、低繁苏尼特羊mRNA相对表达水平差异不显著(P0.05)。
[Abstract]:In order to study the effect of PIWI gene on the development of ovine germ cells, the homologous gene cloning method was used to amplify RNA from testis and ovaries of sheep. The semi-quantitative PIWI gene family was amplified by PCR in Sunita sheep. The results showed that the relative expression level of mRNA in the three members of the PIWI protein family of male sheep was significantly higher than that of female sheep P0.01PIWI2PIWI2PIWI4, and only weak expression of PIWI2PIWI4 was found in the high and small tail Han sheep, and in the low breed Sunita ewe, the expression level of mRNA was significantly higher than that of P0.01PIWI2 and PIWI2PIWI4. There was no significant difference in the relative expression level of mRNA between small tail Han sheep and low procreation Sunita sheep.
