宫颈癌介入治疗前后敏感相关基因的表达谱研究

【摘要】:目的探讨宫颈癌介入治疗前后敏感相关基因的表达谱,分析宫颈癌介入治疗分子机制。方法 3例接受宫颈癌介入治疗患者,治疗前后对其组织标本进行采集,采取基因表达谱芯片对差异表达基因进行筛查,运用分子功能注释系统(MAS)软件对差异表达基因自身功能通路进行挖掘。结果通过对所提取的总RNA电泳图谱进行分析,显示有28S、5S以及18S条带,表示所提取RNA具有较高纯度,能在芯片实验中使用。差异基因共筛选出1162个,和介入治疗前相比,治疗后939个下调基因,223个上调基因。基因功能分析显示涉及的主要有脱氧核糖核酸酶(DNA)复制修复、BRCA1、ATR及BRCA2基因相关通路、细胞有丝分裂、糖酵解代谢路径以及细胞周期调控等。结论表达谱芯片能迅速高通量的进行筛查,且和介入化疗栓塞敏感性有关的差异表达基因具有一定相关性。
[Abstract]:Objective to investigate the expression profile of sensitive genes before and after interventional therapy for cervical cancer, and to analyze the molecular mechanism of interventional therapy of cervical cancer. Methods three patients with cervical cancer received interventional therapy and their tissue specimens were collected before and after treatment. The differentially expressed genes were screened by gene expression microarray, and the functional pathways of differentially expressed genes were mined by molecular function annotation system (MAS) software. Results the total RNA electrophoretic patterns were analyzed. There were 28s 5S and 18s bands, indicating that the extracted RNA was of high purity and could be used in microarray experiments. After treatment, there were 939 down-regulated genes and 223 up-regulated genes. Functional analysis of genes showed that the main genes involved were DNA replication and repair of BRCA1, ATR and BRCA2 gene related pathways, and the cells were mitotic. Conclusion expression microarray can be quickly and high-throughput screening, and the differentially expressed genes related to chemoembolization sensitivity of interventional chemotherapy have some correlation.
