中国荷斯坦牛CXCR1基因SNPs与白血病易感性的关联分析

【摘要】:牛白血病是由牛白血病病毒(Bovine leukemia virus,BLV)引起的牛(Bos taurus)、绵羊(Ovis aries)等动物的慢性肿瘤性疾病,是影响养牛业发展的重要传染病之一。为探索牛趋化因子受体1(chemokine(CX-C motif)receptor 1,CXCR1)基因编码区(coding sequence,CDS)SNPs与牛白血病易感性之间的关系,本研究用荧光共振能量转移-定量多聚酶链式反应(fluorescence resonance energy transfer-quantitative polymerase chain reaction,FRET-qPCR)对某大型奶牛场866头中国荷斯坦牛牛白血病感染情况进行检测,同时用飞行时间质谱法(matrix-assisted laser desorption/ionization time of flight mass spectrometry,MALDI-TOF-MS)对CXCR1基因编码区CXCR1-c.642 AG、CXCR1-c.816CA、CXCR1-c.980AG和CXCR1-c.1068GA 4个SNPs位点进行检测,采用Logistic回归分析CXCR1 SNPs基因型与牛白血病易感性之间的关系,结果表明:在866头奶牛中,患白血病的牛为42头,阳性率为4.85%;CXCR1-c.642 AG、CXCR1-c.816CA、CXCR1-c.980AG和CXCR1-c.1068GA 4个SNPs位点优势基因型分别为GG、CC、AA和GG型,频率分别为0.580、0.466、0.60和0.658。CXCR1-c.980 AG位点对牛患白血病的相对风险影响达到显著水平(P=0.016)。CXCR1-c.980 AG位点GG型基因患白血病的概率为AA型基因的5.04倍。而CXCR1-c.642 AG、CXCR1-c.816CA、CXCR1-c.1068GA和CXCR1单倍型对牛白血病易感性无显著影响(P0.05)。CXCR1c.980AG位点可作为奶牛白血病抗病育种的分子标记之一,筛选CXCR1-c.980AG位点AA型个体有助于降低奶牛对牛白血病的易感性并提高生产效率。
[Abstract]:Bovine leukemia is a chronic tumor disease in animals such as Bos taurus, sheep and other animals caused by bovine leukemia virus. In order to explore the relationship between the coding sequence of CX-C motif)receptor 1 (CXCR1) gene coding sequence and the susceptibility of bovine leukemia. In this study, fluorescence resonance energy transfer quantitative polymerase chain reaction (resonance energy transfer-quantitative polymerase chain reaction) was used to detect leukemia infection in 866 Chinese Holstein cattle in a large dairy. At the same time, matrix-assisted laser desorption/ionization time of flight mass spectrometrymetric MALDI-TOF-MSwas used to detect the CXCR1 gene coding region CXCR1-c.816CAXCR1-c.980AG and CXCR1-c.1068GA. Logistic regression analysis was used to analyze the relationship between CXCR1 SNPs genotype and susceptibility to bovine leukemia. The positive rate of CXCR1-c.642 AGXCR1-c.816CAG CXCR1-c.980 AG and CXCR1-c.1068GA 4 SNPs loci were GGCCCCAA and GG, respectively. The relative risk of bovine leukemia was significantly affected by the frequencies of 0.580, 0.466, 0.60 and 0.658.CXCR1-c.980 AG, respectively. The probability of GG type leukemia at the GG locus of CXCR1-c 980 AG was 5.04 times higher than that of AA gene. The susceptibility of CXCR1-c.642 AGCXCR1-c.816CAA and CXCR1-c.1068GA and CXCR1 haplotype to bovine leukemia was 5.04 times higher than that of AA genotype. CXCR1 c. 980AG locus has no significant effect on the disease resistance breeding of dairy cattle, and it can be used as one of the molecular markers of disease resistance breeding in dairy cattle. The screening of AA type individuals at CXCR1-c.980AG loci is helpful to reduce the susceptibility to bovine leukemia and to improve the productivity of dairy cows.
