伊犁马胃蝇蛆三期幼虫流行病学及COI基因遗传进化分析

【摘要】:为了解伊犁马体内寄生胃蝇属幼虫的种类及马胃蝇蛆的分类。本试验通过对鉴定出的优势种马胃蝇三期幼虫进行流行病学及COI基因分析,采用邻接法(NJ)构建系统发育树。结果显示,共收集16 755个三期幼虫,总感染率为100%,共发现4种胃蝇属幼虫,混合感染现象严重。鼻胃蝇幼虫为优势种,优势度为53.17%,寄生部位更倾向于贲门处,其他3种则无规律地寄生于幽门处。测定的鼻胃蝇蛆、肠胃蝇蛆、兽胃蝇蛆、红尾胃蝇蛆的COI基因序列与GenBank上的序列(登录号:GU265752.1、KR230402.1、KU578262.1、KT946620.1)的核苷酸同源性分别为99%,99%,99%,100%;系统进化分析结果表明,本试验测定与GenBank上发表的胃蝇属的相应蝇蛆种聚类;即肠胃蝇和红尾胃蝇首先聚在一起,再与鼻胃蝇聚在一起,最后三者再和兽胃蝇聚在一起。当以COI序列为靶标时,内群外群分别形成独立的进化分支。该试验结果可为胃蝇属的分类提供参数。
[Abstract]:In order to understand the species of the parasitic larvae of the genus Gastromyza in Yili horse and the classification of the larvae of the genus Musca, this experiment was carried out by epidemiological analysis and COI gene analysis of the third stage larvae of the dominant stallion. Phylogenetic tree was constructed by using NJ). The results showed that 16,755 third stage larvae were collected and the total infection rate was 100. Four species of the genus Gastrozoa larvae were found, and the mixed infection was serious. The larva of the nasal stomach fly was the dominant species. The dominant degree was 53.17, the parasitic site was more inclined to the cardia, the other three species were irregular parasitic in the pylorus. The nasal and stomach maggots, the enterogastric fly maggots, and the animal stomach fly maggots were measured. The nucleotide homology of the COI gene sequence of the maggots in the red tail and the sequence on the GenBank (accession number: GU265752.1 + KR230402.1 + KU578262.1 + KT946620.1) was 999999946620.1.The results of phylogenetic analysis showed that the results of phylogenetic analysis showed that this experiment was similar to the corresponding species of Musca published on GenBank. That is, the stomach fly and the red tail fly first get together, then the nose stomach fly, and the last three get together with the animal stomach fly. When the COI sequence is used as the target, The results of this experiment can provide parameters for the classification of the genus Gypogaster.
