金针菇转运蛋白基因fv-mfs1的序列与表达分析

【摘要】:通过分析金针菇(Flammulina velutipes)子实体原基期、伸长期、成熟期的转录组数据,得到一个差异表达基因fv-msf1。对其结构、序列特征及表达情况进行分析,结果显示该基因全长2709 bp,包含13个外显子,12个内含子,开放阅读框长1770 bp,编码589个氨基酸。结构域分析表明该基因编码的蛋白含主要协助转运蛋白超家族(major facilitator superfamily,MFS)保守结构域。通过实时荧光定量PCR(real-time quantitative fluorescence PCR,q-PCR)对子实体不同发育阶段以及菌盖不同发育时期的表达量进行分析,结果显示该基因在金针菇伸长期的菌盖(尤其是0.3~0.9cm)中高表达,由此推断该基因在金针菇菌盖发育中发挥一定作用。
[Abstract]:A differentially expressed gene fv-msf1 was obtained by analyzing the transcriptional data of Flammulina velutipesae during the primary, extension and maturation stages of the fruiting body of Flammulina velutipesae (Flammulina velutipesae).The results showed that the gene was 2709 BP in length, including 13 exons and 12 introns, and the open reading frame was 1770 BP long, encoding 589 amino acids.Domain analysis showed that the protein encoded by the gene contained major facilitator superfamily (MFS) conserved domain.The expression of Flammulina velutipes (Flammulina velutipes) in different developmental stages of fruiting bodies and different developmental stages of capsid was analyzed by real-time PCR(real-time quantitative fluorescence PCRq-PCRQ. The results showed that the gene was highly expressed in the lid of Flammulina velutipes during the extension period, especially 0.30.9cmof Flammulina velutipes (Flammulina velutipes).It is concluded that this gene plays a certain role in the development of Flammulina velutipes (Flammulina velutipes).
