未控制哮喘与ADRB2基因位点多态性关系的分析

【摘要】:目的:探讨β2肾上腺素能受体(β2-adrenergicreceptor,ADRB2)的基因rs1042718、rs2895795及rs12654778位点基因多态性与未控制哮喘的相关性。方法:选取2015年12月到2016年12月我院确诊为支气管哮规范化治疗半年后未控制者30例为病例组,完全控制及部分控制者30例为阴性对照组,同期我院健康体检者30例为正常对照组,采用Sequenom MassARRAY?SNP检测法和基质辅助激光解吸附电离飞行时间质谱分析质谱技术对三个位点进行多态性检测。结果:rs12654778位点存在GG/GA/AA三种基因型,其基因型的分布频率在三组无统计学意义(GG/GA/AA)且其基因型及等位基因的分布频率在三组间比较差异无统计学意义(P=0.554/0.297/0.819);rs2895795位点TT/AT/AA三种基因型,在三组间比较差异无统计学意义(P=0.105/0.104/0.730);rs1042718位点基因型三组间比较差异无统计学意义(P=0.141/0.051/0.846)。结论:ADRB2基因多态性与未控制哮喘发病无明显相关性,这可能与种族差异、单倍型有关。
[Abstract]:Objective: To investigate the effect of beta 2 adrenergic receptor (beta 2-adrenergicreceptor, ADRB2) genes rs1042718, rs2895795 and the correlation between rs12654778 gene polymorphism and uncontrolled asthma. Methods: from December 2015 to December 2016 in our hospital diagnosed as bronchial asthma standardized treatment after six months without control in 30 cases as the case group, completely control and control part 30 cases were negative control group in our hospital during the same period, 30 healthy subjects served as normal control group, using Sequenom MassARRAY? SNP detection method and matrix assisted laser desorption ionization time-of-flight mass spectrometry mass spectrometry analysis was used to detect the polymorphism of three loci. Results: rs12654778 loci showed GG/GA/AA three genotypes and its genotype the frequency distribution in the three groups was not statistically significant (GG/GA/AA) had no statistical significance and the frequency distribution of genotypes and allele comparison in the differences between the three groups (P=0.554/0.29 7/0.819; rs2895795 TT/AT/AA) loci of three genotypes in three there was no significant difference between groups (P=0.105/0.104/0.730); rs1042718 genotype had no statistically significant difference between the three groups (P=0.141/0.051/0.846). Conclusion: ADRB2 gene polymorphism and uncontrolled asthma had no significant correlation, which may be related to racial differences in haplotype.
