siRNA靶向沉默KIF20A基因对人肝癌SMMC-7721细胞体外增殖和侵袭迁移的影响

【摘要】:目的:研究KIF20A对肝癌SMMC-7721细胞系增殖和侵袭迁移能力的影响。方法:利用RNA干扰技术下调肝癌细胞系SMCC-7721中KIF20A的表达。qRT-PCR和Western blot分别检测细胞KIF20A在mRNA和蛋白水平表达中的变化。CCK-8实验检测增殖能力的改变。Transwell实验检测迁移、侵袭能力的改变。结果:qRT-PCR和Western blot结果显示KIF20A-siRNA能够成功下调KIF20A的表达,CCK-8和Transwell实验分别证实肝癌SMMC-7721细胞增殖和侵袭迁移能力下降。结论:抑制KIF20A的表达能够降低肝癌细胞的增殖和侵袭迁移能力。
[Abstract]:Objective: to study the effect of KIF20A on the proliferation, invasion and migration of hepatocellular carcinoma SMMC-7721 cell line.Methods: the expression of KIF20A in hepatocellular carcinoma cell line SMCC-7721 was down-regulated by RNA interference technique. QRT-PCR and Western blot were used to detect the changes of KIF20A expression at mRNA and protein levels respectively. CCK-8 assay was used to detect the change of proliferation ability. Transwell assay was used to detect migration and invasion ability.Results the results of KIF20A-siRNA and Western blot showed that KIF20A-siRNA could successfully down-regulate the expression of KIF20A. CCK-8 and Transwell experiments confirmed that the proliferation, invasion and migration of SMMC-7721 cells decreased respectively.Conclusion: inhibiting the expression of KIF20A can reduce the proliferation, invasion and migration of hepatoma cells.
