C反应蛋白基因多态性与墨玉县维吾尔族成人慢性牙周炎相关性研究

【摘要】：目的:测定墨玉县维吾尔族慢性牙周炎患者和牙周健康对照者中C反应蛋白+1444C/T rs1130864、+1059G/C rs1800947两位点的基因分型,分析各实验组间基因型分布和等位基因频率的差异;探讨C反应蛋白CRP基因多态性与维吾尔族人群慢性牙周炎易感性的关系,为牙周病的防治提供理论依据和实验基础。方法:将墨玉县维吾尔族人群慢性牙周炎流行病学调查组于前期流调建立的生物样本库作为研究对象,实验分为健康对照组、轻度慢性牙周炎组、中重度慢性牙周炎组,每组随机抽取100例样本,提取DNA,采用聚合酶链反应-限制性片段长度多态性法测定rs1800947,rs1130864两位点的基因分型,统计分析慢性牙周炎的发病是否与C-反应蛋白基因多态性相关。结果:C反应蛋白rs1800947和rs1130864两多态性位点的基因型以及等位基因频率在3组间差异无统计学意义(P0.05);有序多分类Logistic回归分析结果:年龄是慢性牙周炎发病的危险因素(P0.001),性别与慢性牙周炎发病无显著相关(P0.05)。结论:C反应蛋白+1444C/T rs1130864、+1059G/C rs1800947两位点基因多态性与墨玉县维吾尔族成人慢性牙周炎无显著相关性,但有必要增加样本量进行进一步的探索;年龄是患慢性牙周炎的危险因素,且年龄越大越易患慢性牙周炎。
[Abstract]:Objective: to determine the genotyping of C-reactive protein 1444C/T rs1130864 and 1059G/C rs1800947 in Uygur patients with chronic periodontitis and healthy controls in Moyu County, and to analyze the difference of genotype distribution and allele frequency among the two groups. To investigate the relationship between C-reactive protein (CRP) gene polymorphism and susceptibility to chronic periodontitis in Uygur population, and to provide theoretical basis and experimental basis for the prevention and treatment of periodontal disease. Methods: the biological sample bank of chronic periodontitis in Uygur population of Moyu County was divided into healthy control group, mild chronic periodontitis group and moderate and severe chronic periodontitis group. DNA was extracted from 100 random samples in each group. The genotyping of rs1800947 rs1130864 was determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Whether the incidence of chronic periodontitis was associated with C-reactive protein gene polymorphism was statistically analyzed. Results the genotypes and alleles frequency of rs1800947 and rs1130864 were not significantly different among the three groups, and the results of Logistic regression analysis showed that age was the risk factor of chronic periodontitis (P 0.001). There was no significant correlation between sex and chronic periodontitis. Conclusion there is no significant correlation between the polymorphism of 1444C/T rs1130864 and 1059G/C rs1800947 gene and the chronic periodontitis of Uygur adults in Moyu County, but it is necessary to increase the sample size, and age is the risk factor of chronic periodontitis. And the older the more prone to chronic periodontitis.
