旱柳在Pb、Cd胁迫下实时荧光定量PCR内参基因的筛选

【摘要】:为筛选适用于Pb与Cd胁迫下旱柳实时荧光定量PCR的内参基因,应用荧光定量PCR技术分析UBQ1、TUB6、Actin、EF1α、18SRNA、GAPDH、TUA8和UBQ2这8个内参基因在旱柳根部的表达情况。通过Normfinder、ge Norm和Best Keeper3个软件进行综合分析,发现在Pb与Cd胁迫下表达较为稳定的内参基因均为EF1α、GAPDH和Actin,差别是在Pb胁迫下的旱柳根组织中最为稳定的内参基因是EF1α,而在Cd胁迫下的旱柳根组织应最稳定的内参基因为GAPDH。该研究结果可用于旱柳根组织在Pb、Cd胁迫下基因表达的进一步研究。
[Abstract]:In order to screen the internal reference genes suitable for real-time quantitative PCR of Salix willow under Pb and CD stress, the expression of eight internal reference genes (UBQ1, TUB6, EF1 伪 ~ (18) SRNAD, TUA8 and UBQ2) in the root of Salix davidii was analyzed by fluorescence quantitative PCR. Through the analysis of Normfinderge Norm and Best Keeper3 software, It was found that the stable endogenous reference genes expressed under Pb and CD stress were EF1 伪 GAPDH and Actin. the difference was that the most stable endogenous reference gene was EF1 伪 in root tissue of Salix wilfordii under Pb stress, but the most stable endogenous reference gene was GAPDH in root tissue under CD stress. The results can be used to further study the gene expression in the root tissues of Salix wilfordii under Pb and CD stress.
