嗜酸氧化亚铁硫杆菌中铁氧还蛋白基因fdⅠ和fdⅡ的克隆及其重金属抗性研究

【摘要】:嗜酸氧化亚铁硫杆菌是一种应用性很强的工业微生物菌种,鉴于其较好的重金属抗性,可用于贵金属的浸提、环境污染的治理等领域。铁氧还蛋白作为一种广泛存在并参与嗜酸氧化亚铁硫杆菌新陈代谢过程诸如铁硫簇组装等重要环节的蛋白质,研究其结构和功能对了解嗜酸氧化亚铁硫杆菌的代谢过程、电子传递及重金属抗性具有重要意义。本研究的内容和结果如下:(1)从甘肃省兰州市、白银市和陇南市3个典型矿区采集的样品中分离出四株嗜酸氧化亚铁硫杆菌,一株氧化亚铁钩端螺旋菌。选取生长周期短、菌体富集量大的菌株BYSW1进行后续的研究。首先选取温度、pH值、接种量、能源种类利用单因素实验对其生长条件进行优化。在pH为2,于30℃下接种10%的菌液,BYSW1生长最好,葡萄糖和蛋白胨会明显抑制其生长。(2)在NCBI数据库中找出铁氧还蛋白基因序列(fdⅠ、fdⅡ),在BYSW1中扩增出目的片段,连接到pET-32(a),转化大肠杆菌(Escherichia coli,E.coli)DH5α。自BYSW1扩增出的铁氧还蛋白基因序列(fdⅠ、fdⅡ)与数据库汇总的序列一致性分别为83.92%和85.59%,编码的蛋白与数据库中的序列一致性分别为89.47%和85.59%。(3)重组的质粒转化表达宿主E.coli BL21(DE3)。选取温度、时间、IPTG浓度进行单因素实验,优化重组蛋白的表达条件,并用蛋白质印记法进行验证。用镍离子亲和层析柱对重组蛋白进行纯化。FdⅠ和FdⅡ分别在25℃下加入0.5 mM IPTG诱导6 h、25℃下加入0.3 mM IPTG诱导6 h表达效果最佳。FdⅠ和FdⅡ分别可用100 mM咪唑和50 mM咪唑自亲和柱中洗脱下来。(4)挑取五种重金属(Cd2+、Cu2+、Mn2+、Ni2+、Zn2+),对其重金属抗性进行研究;对重金属(Cu2+和Cd2+)驯化前后的嗜酸氧化亚铁硫杆菌的菌体形态和基因组变化、重组蛋白表达差异、宿主菌体的生长情况进行了研究。结果表明:BYSW1具有较强的重金属抗性,分别在0.04 mol/L Cd2+、0.04 mol/L Cu2+、0.15 mol/L Mn2+、0.08 mol/L Ni2+和0.08 mol/L Zn2+存在时生长良好,但是高浓度(大于0.04 mol/L)的Cu2+和Cd2+会明显抑制BYSW1的生长。Cu2+和Cd2+胁迫后,BYSW1的菌体细胞明显变小,且表面粗糙,但基因组仍然能保持完整。不同浓度、不同种类的重金属胁迫前后,铁氧还蛋白的表达存在显著差异,且Cd2+对嗜酸氧化亚铁硫杆菌生长的影响比Cu2+显著;不同浓度、不同种类的重金属对于重组蛋白的表达宿主菌体生长影响不同,0.25 m M的Cu2+和Cd2+能明显抑制宿主菌体的生长。
[Abstract]:Thiobacillus ferrooxidans acidophilus is a kind of industrial microorganism with strong application. In view of its good resistance to heavy metals, it can be used in the extraction of precious metals and the treatment of environmental pollution. Ferredoxin is a widely existed protein involved in metabolic processes of Thiobacillus ferrooxidans, such as the assembly of iron-sulfur clusters. The structure and function of ferredoxin are studied to understand the metabolic process of Thiobacillus ferrooxidans. Electron transport and heavy metal resistance are important. The contents and results of this study are as follows: (1) four strains of Thiobacillus ferrooxidans and one strain of Heptospira ferrooxidans were isolated from three typical mining areas in Lanzhou, Baiyin and Longnan of Gansu Province. The strain BYSW1 with short growth cycle and high concentration of bacteria was selected for further study. The temperature pH value, inoculation amount and energy type were selected to optimize the growth conditions by single factor experiment. The growth of BYSW1 was best when inoculated at 30 鈩,
