基于matK基因的高良姜及其同属混伪品的分子鉴别

【摘要】:目的:建立基于matK基因的高良姜及其同属混伪品的分子鉴别方法。方法:采用通用引物对高良姜样品matK基因进行PCR扩增和双向测序,并从Gen Bank下载10种同属混伪品的matK基因序列。采用Clustal X、MEGA软件进行序列比对分析、计算遗传距离以及构建聚类树。结果:获得的高良姜及其同属混伪品matK基因序列长度为732 bp,变异位点41个。高良姜种内遗传距离为0.000,与混伪品之间的最小遗传距离为0.003。基于matK序列构建的系统发生树显示高良姜样品聚在一起,能直观地与混伪品区分。结论:matK基因可以有效地鉴别高良姜及其同属混伪品。
[Abstract]:Objective: to establish a molecular identification method for Ginger and its concoctions based on matK gene. Methods: the matK gene of Ginger was amplified by PCR and bidirectional sequencing with universal primers, and 10 matK gene sequences were downloaded from Gen Bank. Sequence alignment analysis, genetic distance calculation and clustering tree construction were carried out by using Clustal Xomega software. Results: the length of matK gene was 732 BP with 41 mutation sites. The intraspecific genetic distance of Ginger was 0.000, and the minimum genetic distance between Ginger and its adulterants was 0.003. The phylogenetic tree constructed based on matK sequence showed that the samples of alpinia officinalis were gathered together and could be directly distinguished from the adulterates. ConclusionThe gene of 1: matk can be used to identify alpinia officinalis and its adulterates.
