PTEN基因及miRNA-21在新疆汉族2型糖尿病中的表达及其相关性研究

【摘要】:目的:运用实时荧光定量PCR(q RT-PCR)技术检测PTEN基因及mi RNA-21的表达水平,观察PTEN及mi RNA-21表达水平在新疆汉族2型糖尿病(T2DM)患者与糖耐量正常(NGT)者中的差异,探讨PTEN基因及mi RNA-21在新疆汉族T2DM中的表达及其相关性。方法:选取新疆汉族T2DM患者和NGT者各50例分别作为T2DM组和NGT组;测量并记录一般资料,包括:性别、年龄、身高、体重、腰围、臀围、血压等,计算体质指数(BMI)、腰臀比(WHR);检测空腹血糖(FPG)、餐后2小时血糖(2h PG)、空腹胰岛素(FINS)、空腹C肽、糖化血红蛋白(Hb A1c)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)等生化指标,计算胰岛素抵抗指数(HOMA-IR);应用q RT-PCR技术检测两组PTEN基因及mi RNA-21的表达水平;运用SPSS 20.0软件对数据进行统计学分析。结果:1、新疆汉族T2DM组BMI、WHR较NGT组高,其差异有统计学意义(P0.05);T2DM组性别、年龄、SBP、DBP较NGT组差异无统计学意义(P0.05)。2、新疆汉族T2DM组FPG、2h PG、Hb A1c、ln(HOMA-IR)、TG、TC及LDL-C均高于NGT组,FINS、空腹C肽及HDL-C均低于NGT组,差异均有统计学意义(P0.05)。3、PTEN在新疆汉族T2DM组的表达水平是NGT组的2.18倍。4、mi RNA-21在新疆汉族T2DM组的表达水平是NGT组的2.12倍。5、在新疆汉族T2DM组中,PTEN基因与mi RNA-21的表达无明显相关性(r=-0.078,P=0.592)。结论:1.PTEN基因和mi RNA-21在T2DM中的高表达可能参与了T2DM的发生发展过程。2.在新疆汉族T2DM中,PTEN基因与mi RNA-21的表达可能无关。
[Abstract]:Objective: to detect the expression of PTEN gene and mi RNA-21 by real-time fluorescence quantitative PCR(q RT-PCR, and to observe the difference of PTEN and mi RNA-21 expression in patients with type 2 diabetes mellitus (T2DM) in Xinjiang Han nationality and those with normal glucose tolerance (GTT). To investigate the expression and correlation of PTEN gene and mi RNA-21 in T2DM of Xinjiang Han nationality. Methods: 50 patients with T2DM and 50 patients with NGT in Xinjiang Han nationality were selected as T2DM group and NGT group respectively, and general data were measured and recorded, including gender, age, height, weight, waist circumference, hip circumference, blood pressure, etc. To calculate body mass index (BMI), waist-to-hip ratio (WHR), fasting blood glucose (FPGN), 2 hours postprandial blood glucose (FG), fasting insulin (FINSN), fasting C-peptide, glycosylated hemoglobin (HbA1cN), triglyceride (TGG), total cholesterol (TC), high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), and so on. The expression levels of PTEN gene and mi RNA-21 in two groups were detected by Q RT-PCR technique, and the data were analyzed by SPSS 20.0 software. Results BMIP WHR in Xinjiang Han T2DM group was higher than that in NGT group, the difference was statistically significant (P 0.05). There was no significant difference in age (P 0.05). There was no significant difference between NGT group (P 0.05) and age group (P < 0.05). The TC and LDL-C of T2DM group were significantly higher than those of NGT group (P < 0.05). The fasting C-peptide and HDL-C were lower in T2DM group than in NGT group. There were significant differences in the expression level of P0.05PTEN in T2DM group of Xinjiang Han nationality and 2.18 times of that in T2DM group of NGT group. The expression level of RNA-21 in T2DM group of Xinjiang Han nationality was 2.12 times higher than that in T2DM group of NGT group. There was no significant correlation between the expression of PTEN gene and the expression of mi RNA-21 in T2DM group of Xinjiang Han nationality. Conclusion: 1. The high expression of PTEN gene and mi RNA-21 in T2DM may be involved in the genesis and development of T2DM. The expression of mi RNA-21 may not be related to the expression of PTEN gene in T2DM of Han nationality in Xinjiang.
