橡胶树胶孢炭疽病菌Cg4LysM基因对菌丝生长的影响

【摘要】:本研究通过PT-PCR技术克隆了橡胶树胶孢炭疽病菌的1个候选效应蛋白基因并命名为Cg4LysM。该基因含1 983 bp的完整开放阅读框(ORF),编码660个氨基酸的多肽。生物信息学分析结果表明,该蛋白含有4个LysM保守结构域,不含任何跨膜结构域,其1~20位氨基酸是典型的信号肽序列,推测为胞外分泌蛋白。氨基酸序列比对结果表明,Cg4LysM与草莓胶孢炭疽菌(Colletotrichum gloeosporioides Nara gc5)、高粱炭疽病菌(Colletotrichum sublineola)、石膏样小孢子菌(Microsporum gypseum CBS)的LysM蛋白同源,同源性分别为97%、59%、55%。此外,本研究根据同源重组的原理构建了该基因的敲除突变体△Cg4LysM,进一步分析发现,△Cg4LysM的生长速度慢于野生型菌株,暗示Cg4LysM与胶孢炭疽菌的生长有关。
[Abstract]:In this study, 1 candidate effect protein genes were cloned by PT-PCR technology and named as Cg4LysM., a complete open reading frame (ORF) containing 1983 BP, which encodes a polypeptide of 660 amino acids. The bioinformatics analysis showed that the protein contains 4 LysM conserved domains without any transmembrane domain. 1~20 - bit amino acid is a typical sequence of signal peptide, presumably exocrine protein. Amino acid sequence alignment results show that Cg4LysM is homologous to the LysM protein of strawberry Colletotrichum gloeosporioides Nara gc5, sorghum anthracnose (Colletotrichum sublineola), and Microsporum plaster like Microsporum gypseum CBS. The results were 97%, 59%, and 55%. respectively. In this study, the knockout mutant Delta Cg4LysM of the gene was constructed according to the principle of homologous recombination. Further analysis showed that the growth rate of delta Cg4LysM was slower than that of the wild type, suggesting that Cg4LysM was related to the growth of anthrax spores.
