钙对染铅大鼠海马NMDAR和mGluR5受体介导相关基因mRNA及CaMKⅡ蛋白表达的影响

【摘要】:目的通过研究钙对染铅大鼠海马NMDAR和m Glu R5受体介导相关基因mRNA及CaMKII蛋白表达的影响,为进一步以补钙预防铅的危害提供理论依据。方法选择体质量(50±2)g的清洁级雄性SD大鼠60只,共分五组,每组12只,分别为对照组(基础饲料,蒸馏水)、染铅组(基础饲料,0.2%醋酸铅饮水)、试验Ⅰ组(基础饲料添加0.5%碳酸钙,0.2%醋酸铅饮水)、试验Ⅱ组(基础饲料添加1%碳酸钙,0.2%醋酸铅饮水)和试验Ⅲ组(基础饲料添加2%碳酸钙,0.2%醋酸铅饮水)。连续饲养60 d后,取海马,用荧光定量RT-PCR法测定海马中Grm5、Camk2a、Grin1、Homer1、Prkca和ERK2的mRNA表达量,Western-blotting测定大鼠海马中CaMKII的蛋白表达量。结果与对照组相比,染铅组海马中Grm5、Camk2a、Grin1、Homer1、Prkca、ERK2的mRNA表达量及CaMKII的蛋白表达量均极显著降低(P0.01)。与染铅组相比,试验组钙浓度在一定范围内越高,目的基因表达量相对升高越多,试验Ⅱ组CaMKII的蛋白表达量极显著上升(P0.01)。结论一定浓度的钙可干预铅致大鼠海马组织的损伤。
