广西1例HIV-1G亚型毒株全长基因序列的测定和分析

【摘要】:目的对广西1例HIV-1 G亚型毒株感染者体内病毒近似全长基因组进行序列测定并进行系统进化分析。方法对前期鉴定的1例HIV-1G亚型毒株感染者进行背景信息收集,并采集其外周血,提取病毒RNA,利用逆转录巢式-PCR技术分两段扩增并测定病毒近似全长基因序列。利用MEGA6软件对全长序列进行系统进化分析。结果获得了病毒近似全长基因组序列,长度为8847 bp。在系统进化树中,该毒株与G亚型参考株成簇,Bootstrap值为100%。系统溯源分析显示,该毒株与广西首次报道的G亚型JN106043毒株关系最近(Bootstrap值为100%,基因离散率为5%)。结论该毒株可能是由广西毒株传播所致,提示G亚型毒株在广西地区可能存在一定的局部流行,需进一步监测。该毒株序列的测定和分析对于广西地区HIV流行状况分析具有重要意义。
[Abstract]:Objective to sequence and analyze the approximate full-length genome of a case of HIV-1 G subtype infection in Guangxi. Methods the background information was collected from a case of HIV-1G subtype infection, and the peripheral blood was collected, virus RNAs were extracted, and the approximate full-length gene sequence of the virus was amplified and sequenced by reverse transcription-nested polymerase chain reaction (RT-PCR). The phylogenetic analysis of full-length sequences was carried out by using MEGA6 software. Results the approximate full-length genome sequence of the virus was obtained, the length of which was 8847 BP. In phylogenetic tree, the Bootstrap value of this strain and G subtype reference strain is 100. Systematic traceability analysis showed that the relationship between this strain and the G subtype JN106043 strain reported for the first time in Guangxi was the closest bootstrap value was 100 and the gene dispersion rate was 5%. Conclusion this strain may be caused by the spread of Guangxi strain, suggesting that there may be a local epidemic of subtype G strain in Guangxi, which needs further monitoring. The determination and analysis of the sequence of the strain is of great significance for the analysis of the epidemic status of HIV in Guangxi.
