淀粉酶家族基因在金针菇子实体不同发育时期的表达与分析

【摘要】:为分析淀粉酶对金针菇(Flammulina velutipes)子实体发育的作用,针对已经鉴定的金针菇淀粉酶家族基因(6个α-淀粉酶和1个γ-淀粉酶),采用定量PCR方法对其在不同发育时期(从原基形成第1天到第12天成熟开伞期)、不同组织部位(菌盖和菌柄)的表达量进行测定,进而分析其表达模式。结果显示:7个淀粉酶基因在菌柄中表达量均高于菌盖;其中,α-Amy-5和γ-Amy-1在菌柄中的表达量变化为先下调再上调,两个基因预测编码蛋白均有信号肽,亚细胞定位在细胞外,推测其可能参与菌柄细胞壁的形态建成;α-Amy-3亚细胞定位在线粒体中,与其它各发育时期相比,其在菌柄快速伸长期表达量达到最高。α-Amy-1和α-Amy-6基因则在金针菇成熟期表达量最高,推测其可能与金针菇菌盖的发育有关。
[Abstract]:In order to study the effect of amylase on the development of Flammulina velutipesi fruiting body of Flammulina velutipes. According to the identified genes of Flammulina velutipes amylase family (6 伪 -amylase and 1 纬 -amylase), the different tissues of Flammulina velutipes (Flammulina velutipes) at different developmental stages (from the first day of primordium formation to the 12th day of maturation) were determined by quantitative PCR method. The expression of the bacterial cap and stipe were measured. And then analyze its expression pattern. The expression of 伪 -Amy-5 and 纬 -Amy-1 was down-regulated and upregulated in the petiole, and the signal peptides were found in the predicted proteins of the two genes, and the subcells were located in the cells outside the cells, and the expression of 伪 -Amy-5 and 纬 -Amy-1 in the suspensor was higher than that in the stem, and the expression of 伪 -Amy-5 and 纬 -Amy-1 was down-regulated and up-regulated. It was speculated that 伪 -Amy-3 subcells were located in mitochondria, and the expression of 伪 -Amy-1 and 伪 -Amy-6 genes was the highest in the maturing period of Flammulina velutipes, compared with other developmental stages, the expression of 伪 -Amy-1 and 伪 -Amy-6 genes was the highest. It may be related to the development of Flammulina velutipes.
