木薯种质W_x基因序列的遗传多态性研究

【摘要】:颗粒结合型淀粉合酶(GBSSI)是植物直链淀粉合成的关键酶,主要由W_x基因编码。本研究选用51份木薯种质资源,设计引物对W_x基因进行PCR扩增后测序。通过对获得的W_x基因序列进行了遗传多态性研究,表明其具有很大的保守性。分析W_x基因的SNP分布特点显示其主要分布在第二内含子和第四内含子之间。通过生物软件分析预测其SNP产生对颗粒结合型淀粉合酶(GBSSI)的物理化学、二级结构和三维结构的影响。本研究了解了不同种质间W_x基因的差异,可为木薯的育种提供参考依据。
[Abstract]:Granular-bound starch synthase (GBSSI) is a key enzyme in amylose synthesis, which is mainly encoded by WSP x gene. In this study, 51 cassava germplasm resources were used to design primers for PCR amplification of WX gene and sequencing. The genetic polymorphisms of the Wapx gene were studied, and the results showed that it was highly conserved. The analysis of SNP distribution of WSX gene showed that it was mainly distributed between the second intron and the fourth intron. The effects of SNP production on the physicochemical secondary and three-dimensional structures of granular-bound starch synthase (GBSSI) were predicted by software analysis. In this study, the differences of WSP x gene among different germplasm were studied, which can provide reference for cassava breeding.
