黄秋葵提取物对2型糖尿病小鼠的降糖作用及与基因TNF-α和IDE的相关性研究

【摘要】:目的研究黄秋葵提取物对高脂饮食和链脲佐菌素(STZ)诱导的2型糖尿病小鼠的降糖作用及与肿瘤坏死因子-α(TNF-α)和胰岛素降解酶(IDE)的相关性。方法采用高脂饲料喂养,并ip STZ制备小鼠2型糖尿病模型,随机分为对照组,模型组,二甲双胍组,黄秋葵醇提物(HQK)高、中、低剂量组,黄秋葵水提物(WQK)高、中、低剂量组,均ig给药。给药40 d后进行空腹血糖、口服葡萄糖糖耐量(OGTT)测定、血清和肝组织中相关生化指标检测,并对肝组织进行HE染色。通过荧光定量PCR探讨黄秋葵提取物降糖活性与TNF-α、IDE基因表达的相关性。结果 HQK高剂量组和WQK中剂量组显著降低小鼠空腹血糖,并改善口服葡萄糖耐受量,有效抑制2型糖尿病小鼠血清中总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)和肝组织中AST、ALT,并抑制肝脏中TNF-α、IDE的m RNA表达量,明显提升血清中高密度脂蛋白胆固醇(HDL-C)。肝组织HE病理观察显示HQK高剂量组和WQK中剂量组对肝细胞有一定的保护作用。结论 HQK和WQK通过降低2型糖尿病模型小鼠的TC、TG、LDL-C,同时升高HDL-C来改善小鼠的肥胖、高血脂症状,对2型糖尿病引起的肝损伤有一定的保护作用,并且降血糖与肝脏保护作用与TNF-α和IDE的基因表达量有一定的相关性。
[Abstract]:Objective to study the hypoglycemic effect of Abelia nigra extract on type 2 diabetic mice induced by high fat diet and streptozotocin (STZ) and its correlation with tumor necrosis factor- 伪 (TNF- 伪) and insulin degrading enzyme (IDE). Methods mice model of type 2 diabetes was established by high fat diet and IP STZ. The mice were randomly divided into control group, model group, metformin group, alcohol extract of Abellium nigra HQK (high, middle, low dose, water extract WQK) of high, middle and low dose groups. Both were given intravenously. Fasting blood glucose (FBG), oral glucose tolerance (OGTT), biochemical indexes in serum and liver tissue were measured after 40 days of administration, and liver tissues were stained with HE. To investigate the correlation between the hypoglycemic activity and the expression of TNF- 伪 IDE gene by fluorescence quantitative PCR. Results HQK high dose group and WQK medium dose group significantly decreased fasting blood glucose and improved oral glucose tolerance in mice. To inhibit the expression of TNF- 伪 IDE mRNA in the liver and the serum total cholesterol of type 2 diabetic mice, such as TCX, TGG, LDL-CX, aspartate aminotransferase (AST), alanine transaminase (alt), and the mRNA expression of TNF- 伪 IDE in the liver, and to inhibit the expression of TNF- 伪 IDE mRNA in the liver, and to inhibit the expression of TNF- 伪 IDE mRNA in the liver, and to inhibit the expression of TNF- 伪 IDE mRNA in the liver. High density lipoprotein cholesterol (HDL-C) in serum was significantly increased. Histopathological observation of liver tissue showed that HQK high dose group and WQK medium dose group had protective effect on hepatocyte. Conclusion HQK and WQK can improve the symptoms of obesity and hyperlipidemia by decreasing the TCG-LDL-Cof type 2 diabetic mice and increasing HDL-C, which has a protective effect on liver injury induced by type 2 diabetes. Hypoglycemia and liver protection were correlated with the gene expression of TNF- 伪 and IDE.
