马尾松PmMYB169基因的克隆及表达分析

【摘要】:MYB基因家族在植物生长发育和应答逆境胁迫中有重要作用.通过RT-PCR和RACE技术从马尾松中克隆获得了PmMYB169全长cDNA序列,分析了其在低磷胁迫下的表达特性.PmMYB169全长为1 407bp,开放阅读框为927bp,编码308个氨基酸.生物信息学分析表明,PmMYB169蛋白序列中存在MYB家族结构区域,属于R2R3类MYB转录因子;同源性分析表明,PmMYB169在MYB家族保守区域上同源性较高,它与北美云杉亲缘关系最近.荧光定量PCR对低磷胁迫下PmMYB169的表达分析结果表明,PmMYB169的表达量随低磷胁迫时间延长呈上调趋势,说明PmMYB169参与了低磷胁迫应答;对不同组织的表达分析发现,PmMYB169为组成型表达,在马尾松根茎叶中均有表达,以叶的表达量最高.
[Abstract]:MYB gene family plays an important role in plant growth and response to stress. The full-length cDNA sequence of PmMYB169 was cloned from Pinus massoniana by RT-PCR and race. The expression characteristics of PmMYB169 under low phosphorus stress were analyzed. The full length of PmMYB169 was 1 407 BP, and the open reading frame was 927 BP, encoding 308 amino acids. Bioinformatics analysis showed that there was MYB family structure region in PmMYB169 protein sequence, which belonged to R2R3 MYB transcription factor, and the homology analysis showed that PmMYB169 had high homology in the conservative region of MYB family, and had the closest relationship with Picea spruce. The expression analysis of PmMYB169 under low phosphorus stress by fluorescence quantitative PCR showed that the expression of PmMYB169 increased with the prolongation of the time of low phosphorus stress, indicating that PmMYB169 was involved in the response to low phosphorus stress, and the expression of PmMYB169 was expressed in different tissues. They were expressed in roots and leaves of Pinus massoniana, and the highest expression was found in leaves.
