基因Ⅰ型猪乙型脑炎灭活疫苗免疫原性评价

【摘要】:为了评估猪基因Ⅰ型乙型脑炎病毒YA1株、CZ株灭活疫苗的免疫原性,通过将YA1株、CZ株乙脑病毒分别在乳鼠脑内连续传代,获得了高病毒含量的鼠脑病毒液。将佐剂ISA206分别与灭活的YA1株、CZ株乙脑病毒液混合,制备成鼠脑灭活苗。将YA1株、CZ株鼠脑灭活苗以及HW1株商品化鼠脑灭活苗分别免疫接种小鼠、豚鼠、仔猪,采用间接ELISA试验、血凝抑制试验、病毒中和试验以及攻毒保护试验分别检测IgG抗体、血凝抑制抗体、中和抗体的的产生情况以及疫苗对小鼠的攻毒保护率。结果显示YA1株、CZ株灭活苗在小鼠和仔猪上产生的IgG抗体、在豚鼠上产生的血凝抑制抗体、在仔猪上产生的中和抗体以及对小鼠的攻毒保护率均高于HW1株。结果表明乙脑病毒YA1株、CZ株灭活苗的免疫原性优于HW1株,其中CZ株灭活苗的免疫原性最优。
[Abstract]:In order to evaluate the immunogenicity of the inactivated vaccine against porcine type I encephalitis virus strain YA1 strain CZ strain of Japanese encephalitis virus was subcultured in the brain of the young mice to obtain the high content of the virus in the brain. The inactivated vaccine was prepared by mixing the adjuvant ISA206 with the inactivated YA1 strain CZ strain Japanese encephalitis virus. Mice, guinea pigs and piglets were inoculated with inactivated brain vaccine of YA1 strain CZ strain and commercial inactivated vaccine HW1 strain respectively. IgG antibody was detected by indirect Elisa test, hemagglutination inhibition test, virus neutralization test and attack virus protection test. Hemagglutination inhibition antibody, neutralizing antibody production and the protective rate of the vaccine to mice. The results showed that the IgG antibody produced by inactivated YA1 strain CZ strain on mice and piglets, hemagglutination inhibitory antibody on guinea pig, neutralizing antibody on piglets and the protective rate against HW1 strain were higher than those of HW1 strain. The results showed that the immunogenicity of inactivated vaccine of Japanese encephalitis virus YA1 strain CZ strain was superior to that of HW1 strain, and the immunogenicity of inactivated vaccine of CZ strain was the best.
