Hp感染对丙肝肝硬化外周血Foxp3、RORγt基因表达的影响

【摘要】：目的观察Hp对丙肝肝硬化患者外周血Foxp3、RORγt基因及相关细胞因子表达的影响,初步探讨Hp感染对丙肝肝硬化患者Treg细胞调控作用的影响。方法选取丙肝肝硬化合并Hp感染患者42例为观察组,未合并Hp感染患者30例为对照组,RT-PCR法测定基因表达,ELISA法检测相关细胞因子含量。结果观察组Fox P3、RORγt基因相对表达水平显著高于对照组;观察组IL-10、IL-6、IL-17、TGF-β1、IFN-γ表达水平高于对照组,IL-2表达水平低于对照组;两组患者Foxp3基因的表达与IL-10、TGF-β水平呈正相关,与IL-2水平呈负相关。结论 Hp可能通过诱导Treg细胞的高表达,促进IL-10、TGF-β1、IL-6、IL-17的高表达,抑制IL-2、IFN-γ表达,造成体内Treg/Th17、Th1/Th2平衡紊乱,促进丙肝肝硬化患者肝细胞炎症及纤维化进程。
[Abstract]:Objective to observe the effect of HP on the expression of Foxp3 ror 纬 t gene and related cytokines in peripheral blood of patients with cirrhosis of hepatitis C, and to explore the effect of HP infection on the regulation of Treg cells in patients with cirrhosis of hepatitis C. Methods 42 patients with hepatitis C cirrhosis complicated with HP infection were selected as observation group and 30 patients without HP infection as control group. Results the relative expression level of ROR 纬 t gene in the observation group was significantly higher than that in the control group, and the expression level of IL-10, IL-6, TGF- 尾 1, IFN- 纬 in the observation group was higher than that in the control group, and the expression of Foxp3 gene was positively correlated with the level of IL-10 TGF- 尾 and negatively correlated with the level of IL-2. Conclusion HP may induce the overexpression of Treg cells, promote the high expression of IL-10 TGF- 尾 1 and IL-6N- 纬, inhibit the expression of IL-2 IFN- 纬, cause the imbalance of Treg-Th17 Th1 / Th2, and promote the progression of hepatocyte inflammation and fibrosis in patients with cirrhosis of hepatitis C.
