乐胃饮对胃癌前病变大鼠模型自噬基因Beclin1的影响

【摘要】:目的:观察乐胃饮对胃癌前病变(PLGC)大鼠胃黏膜中自噬体及其相关基因Beclin1的影响。方法:将60只雄性Wistar大鼠分为正常组,模型组,维酶素组,乐胃饮低、中、高剂量组,采用2%水杨酸钠灌胃、饥饱失常以及自由饮用1-甲基-3-硝基-1-亚硝基胍(MNNG)溶液等综合因素诱发PLGC模型。给药28d后,利用透射电镜观察大鼠胃黏膜细胞自噬体,通过RT-PCR检测自噬基因Beclin1 m RNA的表达。结果:与模型组比较,乐胃饮各剂量组胃黏膜Beclin1 m RNA表达显著升高(P0.05,P0.01),电镜可观察到自噬体。结论:乐胃饮可能通过提高Beclin1的表达,增强PLGC胃黏膜自噬而降低癌变的可能性。
[Abstract]:Aim: to observe the effect of Lue Wei Yin on autophagy and its related gene Beclin1 in gastric mucosa of (PLGC) rats with gastric precancerous lesion. Methods: sixty male Wistar rats were divided into normal group, model group, retinol group, low, middle and high dosage group of lewei Yin, which were perfused with 2% sodium salicylate. The PLGC model was induced by combined factors such as hunger and satiety and free drinking of 1-methyl-3-nitro-1-nitrosoguanidine (MNNG) solution. After 28 days of administration, the autophagy of rat gastric mucosal cells was observed by transmission electron microscope, and the expression of Beclin1 m RNA was detected by RT-PCR. Results: compared with the model group, the expression of Beclin1 m RNA in gastric mucosa of each dose group was significantly higher than that in the model group (P0. 05 and P0. 01). Autophagy was observed under electron microscope. Conclusion: Lue Wei Yin may decrease the possibility of carcinogenesis by increasing the expression of Beclin1 and enhancing PLGC gastric mucosal autophagy.
