蜻蜓凤梨AfGAI基因的克

【摘要】:根据蜻蜓凤梨转录组信息,通过RACE技术(rapid amplification of cDNA ends,RACE)获得蜻蜓凤梨中一个DELLA蛋白的cDNA序列,并将其命名为AfGAI。AfGAI cDNA全长1 960 bp,开放阅读框为1 764 bp,编码587个氨基酸。氨基酸序列分析结果显示,AfGAI蛋白含有DELLA蛋白家族共有的功能结构域:DELLA结构域和GRAS结构域,与菠萝(Ananas comosus)、海枣(Phoenix dactylifera)、油棕(Elaeis guineensis)、巨桉(Eucalyptus grandis)、陆地棉(Gossypium hirsutum)中的GAI同源性分别为90%、73%、73%、65%、59%。进一步构建了35S:AfGAI过表达载体,为解析凤梨科植物开花机理提供理论依据。
[Abstract]:According to the transcriptome information of dragonfly and pineapple, the cDNA sequence of a DELLA protein in dragonfly and pineapple was obtained by rapid amplification of cDNA ends (RACE). The full length of the DELLA protein was 1 960 bp, the open reading frame was 1 764 BP and 587 amino acids were encoded. The amino acid sequence analysis showed that the AFGAI protein contained DELLA eggs. The common functional domains of the Bai family are DELLA and GRAS domains, which are 90%, 73%, 73%, 65% and 59% homologous to GAI in pineapple (Ananas comosus), jujube (Phoenix dactylifera), oil palm (Elaeis guineensis), Eucalyptus grandis, and upland cotton (Gossypium hirsutum). The 35S: AfGAI overexpression vector was constructed to understand the above-mentioned genes. To provide theoretical basis for flowering mechanism of pineapple plants.
