小麦3B短臂染色体抗赤霉病主效QTL区域候选基因的表达

【摘要】:通过研究赤霉病菌接种后的基因表达,筛选小麦赤霉病抗性相关基因。根据前人研究结果,利用FGENESH软件对小麦3B短臂抗赤霉病主效QTL所在的小麦基因组测序获得的重叠群ctg0954核苷酸序列进行基因预测,共预测得到489个基因。以携带该QTL的苏麦3号和未携带该QTL的扬麦15为材料,在小麦花期进行赤霉病菌接种诱导基因表达,接种后0 h、4 h、8 h、12 h、24 h对其中有功能注释的259个预测基因的表达进行半定量分析,获得27个在品种间差异表达的基因。这27个基因的表达分为4类:一直上调表达(Ⅰ型),在接种后的早期上调表达后期下调表达(Ⅱ型),接种后早期下调表达后期上调表达(Ⅲ型),接种后一直下调表达(Ⅳ型)。在差异表达基因中,选择6个基因,进行Real-time PCR分析,结果表明这些基因在接种病原菌后均上调表达,且在苏麦3号和扬麦15号品种间存在基因表达差异,验证了半定量表达结果,可作为与赤霉病抗性相关3B短臂主效QTL的候选基因。
[Abstract]:The genes related to scab resistance in wheat were screened by studying the gene expression after inoculation. According to the results of previous studies, 489 genes were predicted by using FGENESH software to predict the nucleotide sequence of overlapped group ctg0954 obtained from wheat genome sequencing of wheat short arm scab resistant to scab. Sumai 3 carrying QTL and Yangmai 15 without QTL were used as materials to induce gene expression by inoculation with scab at flowering stage of wheat.
