贵州省结核分枝杆菌临床分离株MIRU-VNTR位点基因分型研究

【摘要】:目的了解贵州省结核分枝杆菌临床分离株的数目可变串联重复序列(MIRU-VNTR)基因多态性及近期传播情况。方法收集2014年贵州省结核分枝杆菌临床分离株185株(耐多药菌株57株,全敏感菌株97株),采用MIRU-VNTR 12个位点分析法进行PCR检测,应用NTsys2.10软件对全部基因型数字化结果进行非加权组平均法(UPGMA)聚类分析。结果 185株结核分枝杆菌临床分离株分为5个基因群(Ⅰ,Ⅱ,Ⅲ,Ⅳ,Ⅴ),呈现156个基因型,其中135个为独特基因型,其余50株为16簇,最大簇包含10株菌,最小簇包含2株菌,成簇率为27.03%,近期感染最小估计为18.37%,I群结核分枝杆菌为147株,占79.46%,为优势菌群,在MIRU 12个位点中,26、31位点多态性较高(h0.60),4、10、23、27、39、40位点呈中等程度的多态性(0.3≤h≤0.60),2、16、20、24位点多态性较低(h0.30)。结论贵州省结核分枝杆菌具有基因多态性,其主要的流行群为I群,MIRU26和MIRU31呈高等程度多态性,MIRU2、MIRU20和MIRU24、MIRU27呈低等程度多态性。
[Abstract]:Objective to investigate the number of variable tandem repeat (MIRU-VNTR) gene polymorphism and its recent transmission in clinical isolates of Mycobacterium tuberculosis in Guizhou Province. Methods 185 clinical isolates of Mycobacterium tuberculosis (57 multidrug resistant strains and 97 fully sensitive strains) were collected from Guizhou province in 2014. PCR was detected by MIRU-VNTR 12 locus analysis. NTsys2.10 software was used to analyze all the digitized genotypes by unweighted group average method (UPGMA) cluster analysis.
