‘云香’水仙NAC基因克隆及表达分析

【摘要】:从‘云香’水仙转录组文库中筛选出1条NAC基因序列,采用PCR技术克隆得到其c DNA序列。序列分析表明,该NAC基因包含822 bp的开放阅读框,编码273个氨基酸,分子量30 903.9 u,命名为Nt NAC3153(Gen Bank登录号:KU375569)。理化性质分析和多重序列比对显示,Nt NAC3153蛋白是一个无跨膜区域的亲水性蛋白,在N端具有一段保守结构域。系统进化树分析表明,Nt NAC3153与同为单子叶植物的小果野芭蕉Ma NAC、二穗短柄草Bd NAC和小米Si NAC亲缘关系最近。实时荧光定量PCR分析显示,Nt NAC3153基因能够被ABA、盐胁迫和高温诱导表达。研究表明:Nt NAC3153可能参与‘云香’水仙的抗逆调控机制。
[Abstract]:A NAC gene sequence was screened from the transcriptional library of Narcissus sinensis, and its c DNA sequence was cloned by PCR technique. Sequence analysis showed that the NAC gene contained an open reading frame of 822 bp, encoding 273 amino acids with a molecular weight of 30 903.9 us.It was named Nt NAC3153 (Gen Bank accession number: KU375569). Physicochemical properties and multiple sequence alignment showed that Nt NAC3153 protein was a transmembrane free hydrophilic protein with a conserved domain at the N-terminal. Phylogenetic tree analysis showed that Nt NAC3153 had the closest relationship with Bd NAC and Si NAC of Ma NAC, and Si NAC, both monocotyledonous plants. Real time fluorescence quantitative PCR analysis showed that Nt NAC3153 gene could be expressed under ABA, salt stress and high temperature. It is suggested that Nt NAC3153 may be involved in the stress resistance regulation mechanism of Narcissus sinensis.
