绵羊OAS1基因PCR的优化条件

【摘要】:为了研究绵羊OAS1基因片段的PCR最佳反应条件,试验采集绵羊外周血液从中分离出白细胞,并对OAS1基因片段的PCR扩增条件进行优化,通过对引物量、退火温度、模板c DNA的纯度进行筛选,建立最优反应体系。结果表明:PCR反应的最佳cDNA量为2μL,最佳上、下游引物量为1μL,最佳退火温度为60℃。采用上述条件获得的PCR产物,OAS1基因片段的目的条带清晰、明亮,非特异性产物较少。
[Abstract]:In order to study the optimal PCR reaction conditions of sheep OAS1 gene fragments, the white blood cells were isolated from sheep peripheral blood, and the PCR amplification conditions of OAS1 gene fragments were optimized. The purity of template c DNA was screened and the optimal reaction system was established. The results showed that the optimal cDNA content of PCR reaction was 2 渭 L, the optimal number of upstream and downstream primers was 1 渭 L, and the optimum annealing temperature was 60 鈩,
