大豆PAL2-3基因的克隆及转化研究
[Abstract]:Soybean isoflavones are a kind of secondary metabolites in phenylalanine metabolic pathway. In phenylalanine metabolic pathway, phenylalanine ammonia lyase (PAL) is the key enzyme and rate-limiting enzyme. The previous study of our group found that the relative expression of PAL gene and soybean isoflavone content have obvious synergistic increasing or decreasing trend. It is found that PAL2-3 (XM_003542493) is one of the most important members of PAL gene family. In this experiment, PAL2-3 gene was cloned from soybean, then pCAMBIA3301-GmPAL2-3 plant overexpression vector was constructed, and the recombinant plasmid of pCAMBIA3301-GmPAL2-3 expression vector was transformed into Agrobacterium tumefaciens EHA105. The transformed plants were obtained by Agrobacterium tumefaciens mediated transformation system of soybean cotyledon. The transformed plants were detected by PPT, Bar and fluorescence quantitative PCR. The contents of isoflavones in soybean seeds were determined by transgenic positive plants. The results showed that the expression of PAL2-3 gene in T _ 1 transgenic plants was 11.24 times of that of the control, and the highest total isoflavone content (2 587.63 渭 g g ~ (-1) was 1.6-fold higher than that of the control (1 616.90 渭 g g ~ (-1). Therefore, overexpression of PAL2-3 gene in soybean could increase the content of isoflavones in soybean grains.
