胃癌患者Vav1基因D324Y位点多态性检测的意义

【摘要】：目的分析胃癌患者外周血Vav1基因D324Y位点多态性及白细胞介素-6(IL-6)、肿瘤标志物的关系,对其意义进行探讨。方法选取2013年1月-2015年12月确诊胃癌的患者300例为病例组,同时取100例体检健康者为对照组。化学发光法测定血清肿瘤标志物CA199水平;酶联免疫法(ELISA)检测外周血Vav1及IL-6水平;直接测序技术检测Vav1基因D324Y位点多态性情况。结果病例组外周血中Vav1、CA199、IL-6水平均高于对照组(P0.05)。病例组中Vav1基因D324Y位点TT、GT的基因型频率分别为7.0%、19.3%,高于对照组(0%、5.0%)(P0.05);病例组中T等位基因的频率为16.67%,高于对照组(2.5%)(P0.05)。两因素方差分析结果显示,IL-6、CA199、Vav1的水平与患病状态及基因型均有关系(P0.001)。Logistic回归结果显示TT基因型与胃癌关系密切,是胃癌的独立危险因素(P0.05)。结论 Vav1基因D324Y位点多态性可能是胃癌发生进展的重要影响因素。
[Abstract]:Objective to analyze the relationship between Vav1 gene D324Y polymorphism, interleukin-6 (IL-6) and tumor markers in peripheral blood of patients with gastric cancer. Methods 300 patients with gastric cancer from January 2013 to December 2015 were selected as case group and 100 healthy persons as control group. The levels of serum tumor marker CA199, peripheral blood Vav1 and IL-6 were detected by chemiluminescence assay, and the polymorphism of D324Y locus of Vav1 gene was detected by direct sequencing. Results the level of Vav1,CA199,IL-6 in peripheral blood of the case group was higher than that of the control group (P0.05). The genotypic frequencies of TT,GT at D324Y locus of Vav1 gene in the case group were 7. 0 and 19. 3%, respectively, which were higher than those in the control group (0. 0%) (P0.05). The frequency of T allele in the case group was 16.67%, which was higher than that in the control group (2.5%) (P0.05). Two-factor ANOVA showed that the level of IL-6,CA199,Vav1 was correlated with the status and genotype of gastric cancer (P0.001). Logistic regression analysis showed that TT genotype was closely related to gastric cancer and was an independent risk factor for gastric cancer (P0.05). Conclusion the polymorphism of D 324Y locus of Vav1 gene may be an important factor in the development of gastric cancer.
